HCC usually expresses as early vascular invasion, capsular invasion and metastasis inside and outside the liver, although we can take some intervention studies to the recurrence and metastasis of liver cancer on clinic, such as preoperative percutaneous transcatheter arterial embolization(TAE) or chemoembolization(TACE) postoperative TACE and biological treatment interventions, but the overall effect of these measures is still very limited, the key point lies in the molecular mechanisms of HCC recurrence and metastasis is still not entirely clear to us and we realise less knowledge about the molecular regulatory networks. therefore, the study on the liver cancer recurrence and molecular mechanisms and taking effective measures to prevent and treat the medical profession is the major problems need to be solved.Interferon induced transmembrane protein(IFITM) is a discovered antiviral protein family in the cell recently, which inhibit many with or without coated virus’ replication to take effect on antiviral. At the same time, some recent studies show that IFITM3 plays an important role in migration of a variety of human tumor cells and stem cells, but we have not yet realized the liver cancer occurrence, development,diffusion transfer and the mechanism of IFITM3.Purpose:Investigate the biological function of IFITM3 in hepatoma cells.Methods:1. Develop hepatoma cell lines, using Western-Blot method to detect IFITM3’expres-sion in all hepatoma cell lines and select IFITM3 high expression liver cancer cell lines.2. We construct expression vectors of specific sh RNA for IFITM3 and transfect the expression vectors into hepatoma cell lines of overexpression IFITM3. Screeningthe most effective sh RNA interference clips through Western Blot or RT-PCR.3. We will transfect the most effective sh RNA interference clips into IFITM3 high expression of hepatoma cell lines and compare with the normal expression of IFITM3 hepatoma cell lines, detecting the biological of two groups of liver cancer cells through invasion and migration assays.The results:1. Western blot analysis showed that the highest expression of IFITM3 in hepatoma cell line is Hep G2 cell line.2. We were successfully constructing specific sh RNA expression vectors of IFITM3 and transfecting into Hep G2 cell line.3. Transwell assay shows that the invasion and metastasis ability was significantly weakened after the cell interference.In conclusion:1. IFITM3 gene was high expression in hepatoma cell lines.2. If we reduced the expression of IFITM3 in hepatoma cell line,the invasion and metastasis capacity fade.3.
