Objective:1.To detect the expression of interferon-induced transmembrane protein 3(IFITM3)in hepatitis B 、 hepatocellular carcinoma(HCC),and to evaluate the correlation between expression level of the protein and clinical pathology features.2.To investigate Change from hepatitis B to Liver Cancer : the Role of IFITM3。.Methods:1.using immunohistochemistry and Western blot to detect IFITM3 protein expression in hepatocellular carcinoma and corresponding adjacent normal tissues and reveal the relationship IFITM3 expression with liver cancer.2.collected healthy people,hepatitis B virus carriers,liver cancer patient without hepatitis B,liver cancer patient with hepatitis B blood samples as research material,detected protein levels and m RNA expression levels of IFITM3 and confirm whether differences expression among different groups IFITM3,revealed IFITM3 with hepatitis B HCC relationship and to further explore the possibility of its evolution as hepatitis and hepatitis liver cancer early detection biomarkers.3.the area under the ROC curve was screened to detect plasma IFITM3 diagnosis of hepatitis,liver cancer sensitivity and specificity.Results:1.Detected by immunohistochemistry,HCC IFITM3 protein expression was75.0%(45/60),and the adjacent tissues was 11.6%(7/60).2.Western blot detected 60 cases of hepatocellular carcinoma in patients with hepatocellular carcinoma and IFITM3 protein expression in noncancerous tissues:found that with respect to adjacent tissue,liver tissue IFITM3 protein expression was significantly higher,the result was significantly(P <0.05).3.Western blot and QRT-PCR methods to detect three different groups of blood samples found that the expression level of IFITM3: IFITM3 expression levels in the normal population,the patients carry the hepatitis B virus,liver crowd expression level was gradually increase,the difference was statistically significant(P <0.05)Conclusion:1.By hepatocellular carcinoma and adjacent tissues IFITM3 protein expression detection,IFITM3 expression has played a role in the progression of HCC.2.Adoption of healthy people,hepatitis B virus carriers population,HCC crowd IFITM3 plasma protein and m RNA Levels,IFIMT3 expression level was gradually increased,indicating IFITM3 in hepatitis-liver cancer may play a role in the pathogenesis of.3.Adoption of detection of different groups of population serum levels IFITM3,ROC curve analysis showed a greater AUC values,may have an important role in the early detection of early prevention of primary liver cancer,and paly as liver cancer early detection biomarkers.
