Objective:In recent years, ambient mass spectrometry(AMS) is a rapid analytical method which based on a different composition or content molecules in the sample achieved rapid development, and AMS can be in the situation without sample pretreatment for each kind of clinical samples were analyzed. Tip-spray ionization(TSI) is an ion source of the AMS, this study TSI-MS analysis of lung tissues and lung cancer, lung cancer and explore new molecular markers, so as to achieve rapid diagnosis of lung cancer. Methods:With the homemade methanol extraction auxiliary point discharge ion source,we used mass spectrometry to distingues lung cancer and adjacent tissues of 20 lung cancer patients and 9 benign lung disease tissue patients, used the master component analysis(principal component analysis, PCA) method to rapid classificate and identificate the mass fingerprint spectra of lung cancer, adjacent tissues and benign lung disease tissue,and all sample were not be pretreated. The differences m / z 756.7,782.7,808.7,832.7 and standards [DPPC + Na +], [POPC + Na +], [DOPC + Na +], [SAPC + Na +] of 11 patients’ lung cancer and lung cancer adjacent tissues were analized by multistage mass spectrometry. The flow extraction solvent methanol was 4ul / min, and we setted the ion source to the positive ion mode, m / z 50-1000 of mass scan range, m / z 50-1000 of multi-level spectrum scanning range, 150 ℃ of ion transfer tube temperature, 3.5 KV of spray voltage, 65.0 V of lens voltage, 1.6 Da of selected ion isolation width, 13% of the collision energy. Results:By the analysis of anTSI-MS detection and PCA, we can effectively distinguish lung cancer, adjacent tissues, benign lung disease and lung cancer tissues, but the adjacent tissues and benign lung disease tissue can not be distinguished. The multistage average is consistent with the multistage spectrum of standard[DPPC + Na +], [POPC + Na +], [DOPC + Na +], [SAPC + Na +], that is the multistage spectrum of differences in substance m/z spectrum 756.7, 782.7, 808.7, 832.7 in adjacent lung tissue and cancer tissue, i.e. differences in lung cancer substance is a phospholipid DPPC, POPC, DOPC, SAPC. Conclusion:AMS coupling PCA can effectively lung cancer and lung cancer adjacent tissues and benign lung disease tissue by mass fingerprint information,so we can quickly identify lung cancer. TSI-MS multi-stage mass spectrometry can be good for lung cancer and adjacent tissues qualitative differences phospholipid, and DPPC, POPC, DOPC, SAPC may become a new lung cancer molecular markers to provide a new method for the diagnosis of lung cancer.
