Objective:To establish the bone implant in the air pouch mouse model of titanium-particle induced osteolysis, and observe the effect of local injection of titanium ion versus titanium particles sustained on the effect of titanium particle-induced aseptic inflammation and osteolysis. To investigate the titanium ion significance in the process of aseptic loosening of artificial joints.Methods: 60 male Kunming strain mice(aged 8-10 weeks, weighted 30g) were selected. 20 mice selected as the donor of bone graft, the other 40 mice were equally divided into control group(Group A), titanium particle group(Group B), rh-VEGF enhance group(Group C) and titanium ion group(Group D). Air pouches were established on the back of 40 mice by injection 3ml sterile air subcutaneously firstly and the air pouches were added with 1ml of air on alternate days.Results: All mice survived to the end of experiment. The gross observation showed that the red, swollen and vascular proliferation of air pouches were slight in group A, with barely observed new vessel. Group B second to group C, and group D were situated between group B and group C. The histological and molecular biological analysis showed that inflammatory reaction and osteolysis were significantly in group B and the thickness of pouch, cell density and the expression of inflammtory cytokine included TNF-α、IL-1β、IL-6、VEGF were significantly higher than group A(P< 0.05). There are some inflammatory reaction and osteolysis in group C, and higher than group B(P<0.05). Group D observed the same inflammation reaction and lightly osteolysis, but equal or lower than group C(P< 0.05),and higher than group B(P>0.05).Conclusion: Titanium ion liquid could induced inflammatory factor expression in vivo, with the same effect to induce VEGF expression, as the same for the other inflammation factors. Titanium ion could also strengthen the generation and expression of IL-6 and IL-1β.
