Objective: To analyze the correlation and clinical significance of Wnt2 and APC in hepatocellular carcinoma and para-carcinoma tissue,and the exp ression of Wnt2 and APC in hepatocellular carcinoma tissue with different clincopathologic parameters was studied.Methods: Formalin- fixed and paraffine-embedded specimen of 68 cases of HCC during the period from January 2011 to December 2013 were collected from the First Affiliated Hospital of Nanchang University. The experiment was divided into two groups, namely experimental group(group HCC) and control group(group carcinoma tissues).The level of expression of APC and Wnt2 in those three tissues were detected by SP immunohistochemical method, and the data were analyzed statiscally based on clinical pathological parameters.Results:1. Wnt2 mainly expressed in cytoplasm, and partly in nucleus. the rate of expression of Wnt2 in hepatocellular carcinoma tissue, para-carcinoma tissue were respectively 60.3%(41/68), 22.1%(15/68). the expression of Wnt2 between HCC group and para-carcinoma group has statistical significance(P<0.05).the expression of Wnt2 in HCC group were closely related to histological grades, TNM staging(P<0.05).but not related to ages, sexuality, AFP level, tumor size, liver cirrhosis, hepatitis B virus infection(P>0.05).2. APC protein mainly expressed in cytoplasm. the expression rate of APC were 36.8%(25/68)、86.8%(59/68).the expression of APC between HCC group and para-carcinoma group has statistical significance(P<0.05).the expression of APC in HCC group were closely related to histological grades, TNM staging(P<0.05).but not related to ages, sexuality, AFP level, tumor size, liver cirrhosis, hepatitis B virus infection(P>0.05).3. The expression of Wnt2 and APC in HCC showed negative correlation through Spearman correlation analysis(r=-0.43, P<0.01).Conclusions:1. The positive expression rate of Wnt2 increased significantly in HCC compared with para-carcinoma tissue. And it has ne gative correlation with high degree of differentiation, carcinoma in situ. suggesting that it processed the occurrence and metastasis of HCC.2. The expression rate of APC was lower than para-carcinoma, its expression rate decreased significantly in HCC with high degree of differentiation, carcinoma in situ. Indicating that it inhibited the occurrence and metastasis of HCC.3. The negative correlation between Wnt2 and APC showed that APC down-regulated expression or deficiency and Wnt2 up-regulated expression happened in HCC. Thus it will provide theoretical basis for predicting prognosis and therapy of HCC through detecting the Wnt2 and APC.
