Parkinson’s disease is a common neurological degenerative diseases, the main disease characteristics is dopaminergic neurons, dopamine release, substantia nigra striatum dysfunction, the purpose of this paper is to conduct Ndfip1 rat substantia nigra pretreatment, confirmed Ndfip1 protective effect of dopaminergic neurons in the treatment of Parkinson’s disease to provide more theoretical basis and guidance.Objective: to study the the protective effects of Ndfip1 to the 6-OHDA leaded damage of dopaminergic neurons.Methods: The experimental animals were randomly divided into four groups: control group animals don’t be deal with, 6-OHDA was injected in medial forebrain bundle of6-OHDA group animals, 6-OHDA+Adv-None group were injected empty virus on substantia nigra, then 6-OHDA was injected in medial forebrain bundle for two spots,6-OHDA+Adv-Ndfip1 group injected Adv-Ndfip1 on substantia nigra, then 6-OHDA was injected in medial forebrain bundle for two spots, using the determination of animal behavior, immunofluorescence, western blotting and HPLC to decete the related indicators.Results: Adv-Ndfip1 can accurately inject into substantia nigra, and can be successful transfect dopaminergic neurons, immunofluorescence and western blotting can detect the high expression of Ndfip1;After 6-OHDA was injected for two 7 days,the animals can become to the Parkinson’s disease model(>210 for half an hour), TH positive cells were significantly less than the control group(P < 0.05), western blotting is able to detect the expression of TH,it was significantly lower than the control group(P < 0.05);there was no statistically significant difference to detect rotate behavior, the number of TH positive cells and the expression of TH of 6-OHDA group and and6-OHDA+Adv-None group; 6-OHDA+Adv-Ndfip1 group and 6- OHDA group and6-OHDA+Adv-None group, the animals rotate less than the other two groups for half an hour(P < 0.05), the number of TH positive cells significantly more than the other two groups(P < 0.05), the expression of TH is significantly higher than the other two groups(P < 0.05); HPLC detection striatal dopamine content of 6-OHDA group obviously lower than the control group, 6-OHDA+Adv-Ndfip1 group is significantly higher than the 6- OHDA group and 6-OHDA+Adv-None group(P < 0.05).Conclusion: by injecting Adv-Ndfip1 to substantia nigra of the rats, it can reduce the injury to dopaminergic neurons, which is induced by 6-OHDA, it confirmed Ndfip1 plays a protective role of dopaminergic neurons.
