Objection To explore the association of NINJ2 gene polymorphism and vascular cognitive impairment.Methods 112 cases ischemic stoke with cognitive impairment( VCI group),108 cases ischemic stoke without cognitive impairment(NCI group) and 113 healthy controls were involved in this study. Genotypes of single nucleotide polymorphisms(SNPs)(rs11833579 and rs12425791) in NINJ2 gene were analysed by polymerase chain reaction fragment length polymorphism(RFLP)method. Results(1)There was no significant difference in genotype or allele among the three groups of rs11833579(x2=1.942,P>0.05;X2=1.343,P>0.05). After adjusting the risk factors,none genotypes in rs11833579 was significantly associated with the VCI or stoke.(2)There was significant difference in genotype among the three groups(X2=27.036,P<0.05),the frequency of A allele was significant difference among the three groups(X2=19.816,P<0.05). Logistic regression mode can used to performed after adjusting by hypertension,diabetes mellitus,HDL,LDL.The AA+AG genotype of rs12425791 was significant associated with VCI and stoke( OR=1.785, 95%CI 1.008-3.159, P<0.05; OR=1.864,95%CI 1.061-3.274,P<0.05).(3) There were no significant differences among the three groups in gender, age, smokers, blood-triglycerides and total cholesterol. The incidence of hypertension and diabetes were higher in the VCI than the control.Compaired with the control,there were significant difference of HDL and LDL in the VCI(P<0.05). Conclusion :1.The polymorphism of rs12425791 is significantly associated with VCI,the effect could be link to the repairtion of injury in the brain. 2. Hypertension, diabetes, the high level of LDL in plasma may be the risk factors of VCI.
