Objective To study the relationship between endothelial nitric oxide synthase (eNOS) gene polymorphism and vascular cognitive impairment-no dementia after ischemic cerebral disease.Methods 180 patients with ischemic cerebral disease were enrolled randomly into this study. Their clinical materials were collected in detail and all of them received neuropsychological assessment, such as MMSE,MoCA,CDR and CSE-D. Based on clinical materials and assessment results, they were divided into two groups:No cognitive impairment (NoCI) and vascular cognitive impairment no-dementia (V-CIND). A total of 100 healthy people from the Health Management Center was recruited as control group (CR). Polymerase chain reaction combined with restrictive enzyme digestion was used to detect the G894T and 4a polymorphisms of eNOS gene in samples.Results There were significant differences in eNOSG894T genotype distribution and T allele frequency among V-CIND,NoCI and CR. Patients with V-CIND showed a significant higher frequency of the T allele compared to the CR(19%vs8.9%,χ2=10.301, P<0.010, OR=1.73), patients with NoCI also showed a significant higher frequency of the T allele compared to the CR (18.75% vs8.9%,χ2=10.731, P<0.010, OR=2.68), there is no difference in frequency of the T allele between V-CIND and NoCI. As to the frequency of the a allele, there were differences among the three groups (V-CIND 14.5%,NoCI 11.88%,CR 9.6%), but there are no statistically significance.Conclusion 1 The G894T mutation of eNOS is related with V-CIND after ischemic cerebral disease, T allele may be the susceptibility gene, and it effects cognitive function by actting on cerebral diseases indirectly.2 The a allele may be irrelevant to V-CIND after ischemic cerebral disease...
