Background and Objective:Head and neck cancer is one of the major cancers which endangers human life, its pathogenesis is not clear,The main pathological type is head and neck squamous cell carcinoma.Laryngeal carcinoma is the most common head and neck cancer, it remains the global burden of disease.Its high incidence is related with various cause of interaction,such as:Bing Lang chewing,smoking,drinking and HPV infection.At present, the treatment of laryngeal cancer was still dominated by surgical operation,leading operation is functional partial laryngectomy, domestic comprehensive hospitalresection rate is 60% ~ 70%, abroad is relatively high, is80%.scholars believe thatearly detection and early treatment of laryngeal cancer can significantly improve the cure rate and 5 year survival rate in laryngeal carcinoma patients, however in recent years,With the deepeningly development of cancer research in molecular layer, in search of higher diagnostic value of tumor markers is very important in this respect.SOCS2(suppressor of cytokine signaling 2,SOCS2) is a protein which earliest found by Yoshimura,it is composed of 197 amino acid residues. Because it contains a SH2 domain and is named CIS(cytokine inducible SH2-containingprotein).It induced avariety of cytokines,its biological effects via the JAK/STAT pathway to inhibit a variety of cytokines, hormones and growth factors.STAT5(signal transducer and activator oftranscription 5,STAT5) acting on specific DNA fragment in the nucleus,by accepting extracellular signal stimulation of growth factor, cell factor and so on,thus regulae the transcription of target genes, affect cell proliferation,differentiation, apoptosis, participates in the occurrence and development oftumor.The current study shows, SOCS2 and STAT5 are closely related to the development of head and neck cancer,in domestic and abroad, the report of the relationship between SOCS2,STAT5 and laryngeal carcinoma is seldom,esp;een SOCS2 and STAT5,the level of significance is 0.05.Results:1.There are 20 cases with positive expression of SOCS2 in 87 cases of laryngeal carcinomatissues, the positive rate was 23.0%; 17 cases exist positive expression of SOCS2 in 18 cases of adjacent normal laryngeal mucosa, the positive rate was94.4%. There are 70 cases with positive expression of STAT5 in 87 cases of laryngeal carcinoma tissues, the positive rate was 80.5%; 6 cases exist positive expression of STAT5 in 18 cases of adjacent normal laryngeal mucosa, the positive rate was 33.3%.2.The expression of SOCS2 are not related to the parameters of sex and age,but related to the pathological grading,clinical type,lymph node metastasis, pathological staging.The expression level of SOCS2 in high differentiation group was higher than that of low differentiation group;the expression level of SOCS2 in no lymph node metastasis group was higher than that in lymph node metastasis group; The expression level of SOCS2 in I-II stage group was higher than that in III-IV stage group,the difference was statistically significant(P < 0.05).3.The expression of STAT5 are not related to the parameters of sex,age,pathological grading,clinical type,pathological staging,but related to lymph node metastasis.The expression level of STAT5 in lymph node metastasis group was higher than that in no lymph node metastasist group,he difference was statistically significant(P <0.05).4.SOCS2 and STAT5 protein have different levels of expression in laryngeal carcinoma,statistical analysis using Spearsman correlation, the results showed a negative correlation between them(P < 0.05).Conclusions:1.The expression of SOCS2 and STAT5 in normal adjacent tissues and laryngeal carcinoma were different, SOCS2 are closely related with the clinical and pathological features of patients with laryngeal cancer(t pathological grading,clinical type,lymph node metastasis, pathological staging), STAT5 are closely related with the clinical and pathological features of patients with laryngeal cancer(lymph node metastasis), both of them might be involved in the occurrence and development of laryngeal carcinoma.It indicates that the two may be as the index and basis of laryngeal cancer progression and prognosis.2.SOCS2 plays an inhibitory role in laryngeal carcinoma progression, as a tumor suppressor gene involved in the infiltration of laryngeal carcinoma; the expression of STAT5 plays an activation role in laryngeal carcinoma occurrence and development,as cancer gene involved in laryngeal carcinoma, It indicates that activation of SOCS2 or inhibition of STAT5 may prevent laryngeal cancer progression or has an effect on the treatment of laryngeal cancer.3.The expression of SOCS2 and STAT5 were negatively correlated, it suggested that earlycombined detection of the expression level of SOCS2 and STAT5 may provide a reliable basis for early laryngeal cancer diagnosis, treatment, prognosis.
