Objective:To investigate the effect on the levels of blood lipid and the expression of phospholipid transfer protein(PLTP) caused by berberine in the early atherosclerosis model of Wistar rats. 1)To make a theoretical basis of PLTP for the clinical diagnosis in atherosclerosis. 2)Through statistical system to evaluate the effect of berberine intervenes PLTP of serum in the early atherosclerosis rats and it’s intervention in atherosclerotic lesions. 3) Evaluate the efficacy and safety of berberine for cardiovascular disease prevention.Method:1、Intraperitoneal injection of vitamin D3 600000 IU / kg and High fat diet to establish the early atherosclerosis model of Wistar rats.2、High dose of berberine 300mg·kg-1·d-1, low dose of berberine 37.5 mg·kg-1·d-1irrigation AS model of Wistar rats.3、The animals were sacrificed, take 5-10 ml blood from abdominal aorta. Using procoagnlant separating gel tubes for making the serum; lift off length of aorta along the aortic valve to the branches of iliac artery aorta.4、Manufacture the serum from the blood, using colorimetry to detect PLTP, VLDL and using ELISA to assay TG, TC, LDL-C, HDL-C,.Result:1 、 The model control group compared with the control group: the expression of TC,TG,LDL-C, HDL, PLTP, VLDL were significantly increased, the difference were statisticallysignificant(P<0.05)2、The low dose group of Berberine(37.5 mg·kg-1·d-1) compared with the model control group:The expression of TC, TG, LDL-C, PLTP, VLDL was significantly decreased, HDL-C was increased slightly. There were statistically significant difference comparison on TC, LDL-C, VLDL,PLTP(P<0.05) but no statistically significant difference comparison on TG,HDL.3 、 The high dose group of Berberine(300 mg·kg-1·d-1) compared with the model control group: The expression of TC, TG, LDL-C, PLTP,VLDL was significantly decreased, HDL-C was increased. There were statistically significant difference comparison on TC, LDL-C,HDL,VLDL, PLTP(P<0.05)but no statistically significant difference comparison on TG.4 、 The high dose group of Berberine(300 mg·kg-1·d-1) compared the low dose group of Berberine(37.5 mg·kg-1·d-1) : The expression of TC, TG, LDL-C, PLTP, VLDL was significantly decreased, HDL-C was increased. There were statistically significant difference comparison on TC,LDL-C,PLTP(P<0.05) but no statistically significant difference comparison on TG, VLDL.5 、 Berberine can reduce the progression of early to intermediate AS in a dose-dependent manner, There was no toxic side effect performance that High dose group and low dose group of berberine gavages for 7weeks.Conclusion:1、Berberine could prevent and treat the progression of early to intermediate AS and showed a significant effect of high doses.2、Berberine can reduce TC, LDL-C, PLTP, VLDL in the serum,but no significant effect on TG.3、 Berberine can be raised the level of HDL when high doses, but compared with the low dose was not statistically significant effect on TG,VLDL.
