Objective: Through the screening of Chinese bladder cancer cell line BIU-87, research team got Chinese small molecular polypeptide CSSPIGRHC in the prophase, the experiment is combined with the identification of the polypeptide Chinese bladder cancer cell line BIU-87 with specificity and strength of binding force.Methods: I developed 4 kinds of cancer cells: Chinese bladder cancer cell line BIU-87, human liver cancer cell line SMMC-7721, human breast cancer line MCF-7, human cervical carcinoma cell line Siha, which combined with fluorescent small molecule polypeptide(fluorescence) FITC-CSSPIGRHC, observation and flow cytometry assay under confocal laser scanning microscope.Results: 1. The experiment has been successful in cultivating Chinese bladder cancer cell line BIU-87, human hepatocellular cancer cell line SMMC-7721, human breast cancer line MCF-7, human cervical cancer cell line Siha, each cell line grew well, and meet the experimental requirements. 2. Confocal laser scanning microscope(CLSM) observation showed that: 2.1 FITC-CSSPIGRHC fluorescent probe can be combined with Chinese bladder cancer BIU-87 cells, human hepatoma SMMC-7721 cells, human breast cancer MCF-7cells and human cervical cancer Siha cells(in green). 2.2 Average fluorescence values of fluorescence probe FITC-CSSPIGRHC has been combined with Chinese bladder cancer BIU-87 cells stronger than in human hepatoma SMMC-7721 cells, human breast cancer MCF-7 cells and human cervical cancer Siha cells. The difference was statistically significant(P < 0.05). 3. Flow cytometry(FCM) detection display: 3.1 FITC-CSSPIGRHC fluorescent probe can be combined with Chinese bladder cancer BIU-87 cells, human hepatoma SMMC-7721 cells, human breast cancer MCF-7 cells and human cervical cancer Siha cells(mean fluorescence intensity value is higher than the blank group). 3.2Average fluorescence intensity value of fluorescence probe FITC-CSSPIGRHC has been combined with Chinese bladder cancer BIU-87 cell is higher than human hepatoma SMMC-7721 cells, human breast cancer MCF-7 cells and human cervical cancer Siha cells, the difference was statistically significant(P < 0.05). 4. Statistical analysis results show: Chinese small molecular polypeptide CSSPIGRHC can combine with bladder cancer cell line BIU-87, and the strong,meanwhile can combine with the other 3 system cancer cells. But the former is more obvious strong than the latter in the aspect of binding force,the difference was statistically significant(P < 0.05).Conclusion: Chinese small molecular polypeptide CSSPIGRHC can specifically bind to Chinese bladder cancer BIU-87 cell specific, it may be used as Chinese bladder cancer BIU-87 cell specific targeting peptide.
