| Objective:Comparison of the difference of the functional connectivity between Alzheimer’s disease(AD) patients, mild cognitive impairment(MCI) patients and normal person who carried Apo Eε4 allele or non-carried,to investigate the influence of Apo Eε4 allele on brain functional status,and to assess the potential value of resting-state functional magnetic resonance imaging and Apo Eε4 allele in cognitive function. And provide valuable non-invasive radiological indicators and biomarkers for the early diagnosis of MCI and AD. Methods:Oral mucosa from 16 AD patients,16 MCI patients and 16 normal person were collected from the First Hospital of Shanxi Medical University in April 2014-February 2015 for Apo Eε4 allele testing.Conventional brain MR examination and resting-state BOLD-f MRI sacans was performed in all people,to analysis the differences of Apo Eε4 allele(chi-square test) and the brain functional connection(Each group carries and noncarriers were compared using t test). Results:1.The AD group, MCI group and the normal control group have no significant difference in age, sex and educational attainment;2.The AD group, MCI group and the normal control group have difference(c2=7.462.P=0.024) in Apo Eε4 genotype expression, Apo Eε4 carring rate in AD group is the most,MCI group is less,and the normal control group is the least.3.Apo Eε4 carriers in the AD group, MCI group and the normal control group compared with the non-carriers,some regions of the brain is reduce in local connection,eg:the left inferior parietal lobule;the left middle occipital gyrus, the right middle occipital gyrus;the right fusiform gyrus,the right insula tegmental area, the right middle occipital gyrus,the right insula; Apo Eε4 carriers in the ADgroup and the normal control group compared with the non-carriers,some regions of the brain is increase in local connection,eg: the right side of supplementary motor area,the right frontal gyrus;the left centralis posterior gyrus,the left thalamus and the left inferior frontal gyrus;4.Apo Eε4 carriers in the AD group, MCI group and the normal control group compared with the non-carriers, the DC value reduced in some regions of the brain,eg:the right superior frontal gyrus,the left anterior cingulate gyrus,the left basal ganglia,the left insula,the left inferior frontal gyrus; the left superior occipitals gyrus,the left,the right superior middle frontal gyrus, the right middle frontal gyrus;the right superior frontal gyrus,the right middle occipitals gyrus,the left superior occipitals gyrus,the left calcarine,the left inferior frontal gyrus,the left lingual gyrus; Apo Eε4 carriers in the AD group, MCI group and the normal control group compared with the non-carriersthe,DC value increased in some regions of the brain,eg:the right frontal gyrus,the right occipital gyrus,the left lingual gyrus,the left parahippocampal gyrus;the right superior frontal gyrus;the left superior temporal gyrus,the left middle temporal gyrus,the right precuneus. Conclusion:1.Apo Eε4 Allele genotype positive in AD patients is higher than the normal elderly;2.the default net work connection of patients who carrying Apo Eε4 Allele is reduced,and have a promoting role in prominent clinical features of AD---episodic memory, emotional integration even language barrier. |
