Objective:To explore the influence of IL-24 in combination with cisplatin on the cervical cancer nude model of lymphatic metastasis, confirmed the completion of lymph node metastasis in cervical cancer cells through the regulation of HMGB1 / RAGE signaling pathways.Methods:Siha cells were seeded in 42 side of the rear paw-pads of nude, the nude model of lymphatic metastasis was successfully established and randomly divided into six groups,each 7. Intervention with PBS, p DC316, recombinant plasmid with IL-24 gene,recombinant plasmid with IL-24 gene plus low-dose cisplatin, low-dose cisplatin,high-dose cisplatin to observe the general condition of nude mice with lymphatic metastasis. To certified internal invasion and metastasis in lymph node with pathological methods, and detecte the expression of IL-24 gene in tumor by real time PCR, and detected the expression of HMGB1 / RAGE signaling pathways by immunofluorescence and westernblot.Results:1.The tumor volume and weight of each group was statistically significant difference in nude before interventions treatment; 2 the number of lymph node metastasis in nude mice between the groups was statistically significant difference after interventions treatment, p DC316-h IL- 24 with low cisplatin group was significantly higher than the other groups; 3. drawn parts of the lymph nodes and cell invasion confirmed by pathology;4.IL-24 gene was success stably transfected in tumor that was detected by real time PCR; 5after intervention treatment on nude mice in each group, the efficiency of fluorescence expression in HMGB1 / RAGE signaling pathways were different, the fluorescence expression of combined group was significantly less than the other groups; 6.after intervention treatment on nude mice, the protein expression of each group in nude lymph of HMGB1 / RAGE signaling pathway was statistically significant difference that was detected by westernblot method, protein expression of combined group was significantly less than the other groups.Conclusions:The group of p DC316IL-24 with cisplatin significantly inhibited lymph node metastasis, the inhibition of lymph node metastasis in cervical cancer cells is completed by HMGB1 / RAGE signaling pathway proteins, it is confirmed that the mechanism of lymph node metastasis in cervical cancer cells and HMGB1 / RAGE signaling pathways are involved, it provide new ideas for achieving the inhibition of lymph node metastasis on cervical cancer.
