| Objective To study the effect of inhalation anesthesia of sevoflurane on cognitive function in the DACD aged rats, and explore its mechanism in role of neurotransmitter through analysis the content of Glu and GABA in hippocampus, and it can also provide the reasonable use of narcotic and anesthesia methods for DACD patients.Methods Ninety cleaning SD rats, were randomly divided into 3 groups: the control group(Group C, n= 30) : fed the rats by using common forage for 8 weeks, then injected the citric acid diluent into the intraperitoneal, after that continued to feed the rats by the original common forage for another 7 weeks, during the period, freely water drank; Diabetes group(Group D, n=30) : fed the rats by using the high sugar and high fat for 8 weeks, then received intraperitoneal injection of streptozotocin(STZ), after that, continued to feed the rats by the original feeding scheme for another 7 weeks, during the period, freely water drank, the purpose was to establish model of type 2 diabetes mellitus(T2DM); The sevoflurane group(Group DS, n=30) :It also followed the scheme in making animal model of the Group D. After modelling of DACD rats was success by Group DS, it was randomly divided three subgroups by the duration of 1.5% sevoflurane inhalation( n=10, respectively): DS1( 1 hour), DS2( 2 hours), DS3(3 hours). The group C and the group D were also randomly divided 3 subgroups by the duration of inhalation of 100% oxygen: C1( 1 hour), C2( 2 hours), C3( 3 hours) and D1( 1 hour), D2( 2 hours), D3( 3 hours). After 1 day and 5 day of dealing with the rats by inhalation anesthesia of 1.5% sevoflurane or 100% oxygen, we compared the level of cognitive function of the rats which were handled with the rats which were not. And then extracted the CA1 area tissue of hippocampal, to determine the content of Glu and GABA and the ratio of Glu/GABA.Results 1, Fed by high sugar and fat composite STZ was injected into intraperitoneal can successfully established DACD rat model. 2, Compared with the level of cognitive function of the subgroup in group C which was not treated, each subgroup in group D and group DS was reduced, the differences were statistically significant(P<0.05); Compared with C3, D3 about the level of cognitive function after 1 day of the rats were treated, DS3 was statistically significant reduced, the differences was statistically significant(P<0.05); 3, Compared with C3, D3 about the Glu, GABA and the ratio of Glu/GABA after 1 day of the rats were treated, the content of Glu was reduced, but the GABA was rose, and the ratio of Glu/GABA were reduced in the group DS3, the difference were statistically significant(P< 0.05);Conclusion Inhalation anesthesia of 1.5% sevoflurane more than 3 hours could induce short term cognitive impairment in the DACD aged rats, the mechanisms might associate with the content of Glu decreased in hippocampus CA1 area, while increased the content of GABA. |
