Objective To test the expression of programmed death-1(PD-1),programmed death ligand-1(PD-L1) and tumor necrosis factor-alpha(TNF-alpha) in cholesteatoma otitis media, explore the role of PD-1,PD-L1 and TNF-alpha in cholesteatoma otitis media’s proliferation and invasion.Methods To test the expression of PD-1,PD-L1 and TNF-alpha in 39 cases of cholesteatoma otitis media tissues and 15 cases of normal skin of external auditory canal by immunohistochemical technology(PV-9000).Result The percentage of PD-1,PD-L1 and TNF-alpha expression in cholesteatoma otitis media was higher than that in normal skin of external auditory canal(χ2=31.949,P﹤0.05;χ2=15.815,P﹤0.05;χ2=27.662,P﹤0.05).The expression of PD-1,PD-L1 and TNF-alpha in cholesteatoma otitis media had positive correlation with the degree of inflammation(r=0.456,P=0.004; r=0.614,P=0.000; r=0.737,P=0.000).In cholesteatoma otitis media, the expression of PD-1 had positive correlation with that of PD-L1(r=0.510,P<0.01), the expression of PD-L1 had positive correlation with that of TNF-alpha(r=0.392,P<0.05), the expression of PD-1 had positive correlation with that of TNF-alpha(r=0.636,P<0.01).Conclusions The percentage of PD-1,PD-L1 and TNF-alpha expression in cholesteatoma otitis media was higher than that in normal skin of external auditory canal. The expression of PD-1, PD-L1 and TNF-alpha in cholesteatoma otitis media had positive correlation with the degree of inflammation. TNF-alpha has crucial importance in the process of bone damage in cholesteatoma otitis media. TNF-alpha and PD-1/PD-L1 signaling pathway have synergetic effects in the proliferation and invasion of cholesteatoma otitis media.
