ObjectiveThis paper is to investigate the effect of FK1706 on the behavior change of the T8 spinal cord injured rats, and to discuss the possibility of FK1706 on treatment of spinal cord injury.Methods55 SD male rats, 200-250 g, were randomly divided to A(the Sham-operation group), B(the SCI group), C(the MP group), D(the FK1706 group) and the E(Combination group). Every group has 11 rats. The modified Allen’s construction was used to build a model by hitting the T8 spinal cord. And then 3 rats for SEP monitoring were randomly selected. B, C, D and E groups were all consistent in morphological change, and the 3 rats were all consistent in SEP monitoring, which meant the building model was successful.After the model was completed, methylprednisolone(40mg/kg)was injected into the tail vein of the rats in the C group, the same volume of physiological saline was injected into the rats in the A, B and D group at the same time. FK1706(1mg/kg)was injected into the tail vein of the rats in the D group, the same volume of physiological saline was injected into the rats in the A, B and C group at the same time. methylprednisolone(40mg/kg)and FK1706(1mg/kg)were injected into the tail vein of the rats in the E group. After the intervention, execute rats and collect the samples in the T8 spinal cord for real-time PCR reaction to detect the expression of IL-1β.After the model was completed, methylprednisolone(40mg/kg)was injected into the tail vein of the rats in the C group for 35 days, the same volume of physiological saline was injected into the rats in the A, B and D group. FK1706(1mg/kg)was injected into the tail vein of the rats in the D group for 35 days, the same volume of physiological saline was injected into the rats in the A, B and Cgroup. methylprednisolone(40mg/kg)and FK1706(1mg/kg)were injected into the tail vein of the rats in the E group for 35 days. The BBB score and the inclined plane test were used to estimate the function of hind limbs behavior at the 3rd days, 1st week, 2nd week, 3rd week, 4th week and 5th week after the surgery. These tests were used to investigate the behavior change of the T8 injured rats that were treated with drugs.ResultsPart one: The expression of IL-1β in the Sham-operation group is different with other groups(P<0.05), there is no difference between the FK1706 group and the SCI group(P>0.05.), there is significant difference between the FK1706 group and the MP group(P<0.05), there is no difference between the MP group and the Combination group(P>0.05).Part two: 1 rat died in B group, 2 rats died in C group. BBB score result: there is no significant difference in all groups except the Sham-operation group(p>0.05). The MP group and the Combination group are different with the SCI group(p<0.05)respectively at the 1st week. The MP group, the FK1706 group and the Combination group are different with the SCI group(p<0.05)at the 2nd week, 3rd week, 4th week and 5th week respectively. at the 4th week, there is difference between the MP group and the SCI group(p<0.05),there is difference between the MP group and the FK1706 group, there is difference between the MP group, the FK1706 group and the combination group(p<0.05). The inclined plane score: The MP group and the Combination group have no difference with the SCI group(p>0.05)at the 3rd day and 7th day. There is difference between the MP group and the Combination group from 1st week to 5th week. There is no difference between the MP group and the combination group at 3rd day and 7th day(p>0.05).There is difference between the MP group and the combination group from 2nd week to 5th week(p<0.05). There is difference between the MP group and the FK1706 group from 3rd day to 2nd week(p<0.05).There is no difference between the MP group and the FK1706 group at 3th week(p>0.05). There is difference between the MP group and the FK1706 group from 3th week to 5thweek(p<0.05). There is difference between the MP group and the Combination group at all times(p<0.05).Conclude1 、 Methylprednisolone can depress the expression of IL-1β, FK1706 has few inhibiting effect on IL-1β, the Combination group only show the effect of methylprednisolone, without coordinate repression of IL-1β. 2 、 The behavior score of rats treated with FK1706 is better than methylprednisolone, showing that FK1706 can promote the nerve growth. 3 、 The score of spinal cord injury in rats in the combination group(methylprednisolone+FK1706) is higher than in the MP group or the FK1706 group, which proves that methylprednisolone and FK1706 have mutual promoted effects in treatment.
