Objective: Observed anterior cruciate ligament transection( ACLT) to build clinical pathological features of SD rat model of osteoarthritis(OA), and to examine the expression of mi R-146 a, mi R-27 a and u PA in joint cartilage and synovial of SD rat model of OA.Methods: 120 SD rats were divided into experimental group and control group. Experimental group received unilateral ACLT, control group the treatment was similar to the experrnental groups,but no transction ACL, and they were sacrificed at 2, 4 and 8 weeks after surgery respectively, knees were harvested to observe the pathologic change of cartilage macroscopically and histologically, histopathological grading OA was performed according to a Mankin’s scoring system. The expression level of mi R-146 a, mi R-27 a and u PA in joint cartilage and synovial tissue were detected by real-time fluorescent quantitative polymerase chain reaction.Result:1.Two weeks after ACLT cartilage surface roughness and color gloomy were seen, and become more severe at 4 and 8 weeks after operation, while cartilage still remained normal after pseudo operation. The experimental group different time periods Mankin score significant differences with the control group.2. In the experimental group and control group, the relative expression level of mi R-146 a at2,4,8 weeks in joint cartilage were(0.58±0.13)vs(0.66±0.14)、(0.43±0.08) vs(0.68±0.12)、(0.18±0.06)vs(0.61±0.07), the relative expression level of mi R-27 a at 2,4,8 weeks in joint cartilage were(0.48±0.15) vs(0.58±0.16) 、(0.33±0.08) vs(0.59±0.09) 、(0.13±0.04) vs(0.6±0.08), the relative expression level of u PA at 2,4,8 weeks in joint cartilage were(0.32±0.05) vs(0.41±0.03)、(0.34±0.06) vs(0.88±0.09)、(0.31±0.04) vs(1.18±0.07), each time point where statistically significant differences(P<0.05). The expression level of mi R-146 a,mi R-27 a and u PA in synovial tissue had significant differences(P<0.01).3. Application pearson correlation analysis showed that the expression level of mi R-146 a,mi R-27 a and u PA in cartilage tissue were significantly correlated to Mankin score(P<0.05),the expression level of mi R-146 a and mi R-27 a were reduced as the cartilage tissue degradation, but the expression level of u PA was increased. In the degradation of synovial tissue, the expression of mi R-146 a, mi R-27 a and u PA were significantly correlated to each other.Conclusion:1. Anterior cruciate ligament transection(ACLT)to build clinical pathological features of SD rat model of osteoarthritis(OA), the pathological changes of OA were comprehensively observed in the whole process. This method was applied for simple technology, but had less surgical trauma, and it was suitable for the study of osteoarthritis because of a good human biological similarity.2. In the process of degradation, the low expression level of mi R-146 a and mi R-27 a were3. significantly negatively correlated to the high expression level of u PA in the cartilage and synovial tissue.4. To detect the expression level of mi R-146 a, mi R-27 a and uPA in the cartilage and synovial tissue and analyze the relationship between them would further clarify the pathogenesis of osteoarthritis. At the same time, the research provided new theoretical and experimental basis for the targeted gene therapy of osteoarthritis.
