Objective The aim of this study is to investigate the effect of CBS overexpression on cognition and APP amyloidogenic pathway changes in APP/PS1 transgenic mouse.Methods CBS overexpression lentivirus were injected into the APP/PS1 transgenic mice hippocampus, dummy vector lentivirus as negative control. After 4 weeks, cognitive function were measured by Morris water maze, Western Blot detect BACE-1、PS1 and the expression and distribution of Aβ1-42 were detected through Elisa and immunohistochemical staining.Results ①Lentivirus vector which overexpress CBS can increase the content of H2S level effectively (P<0.05); ②Overexpress CBS can improve the learning and memory ability of transgenic mice, lead to the escape latency shorted and the crossing plat times increased (P<0.05); ③The expression of BACE-1,PS 1 is reduced and the level of Aβ1-42 is decreased in CBS-overexpression group(P<0.01)Conclusion CBS overexpression can ameliorate the APP/PS1 transgenic mouse cognitive dysfounction, inhibit amyloidgenic pathway key enzymes and reduce the generation of Aβ, this result could be achieved by increased H2S indirectly.
