| Objective:Women with polycystic ovarian syndrome (PCOS) mostly associated with different levels of insulin resistance (IR). Although its reported that Irisin has certain correlation with IR in type 2 diabetes, the relationship of serum Irisin levels and IR in women with PCOS are still not available. Our study was to investigate the serum Irisin level in patients with polycystic ovary syndrome and controls; and the relationship between the serum Irisn and IR and the metabolic parameters associated with glucose and lipid metabolism.Methods:164 newly diagnosed patients with PCOS and 126 healthy age-matched controls are recruited in this study. All subjects were measured height, weight, blood pressure, waist circumference, hip circumference, waist-to-hip ratio and BMI were calculated, Blood lipids, hormones and OGTT were measured in Clinical laboratory. Euglycemic hyperinsulinemic clamp (EHC) were performed to evaluate the insulin sensitivity (insulin resistance), represented by M value. Irisin serum concentration was measured by Enzyme-linked immunoassay.Results:(1) PCOS patients had higher BMIã€WHR. FAT%ã€SBPã€DBPã€TG〠TCã€HDL-Cã€LDL-Cã€FBGã€2hPBGã€FInsã€2hInsã€HbA1cã€TEST. LH/FSHã€PRLã€FAI than controls, and lower SHBG(P<0.05 or P<0.01).(2) Serum level of Irisin are siglificantly higher in women with PCOS(P<0.01); Obesity subgroups (BMI≥23kg/m2) has higher serum Irisin level than non-obesity subgroups in both PCOS patients and controls(P<0.01).(3) In EHC, M value are decreased in PCOS patients (P<0.05). Brief rapid rise in insulin levels could cause the concentration of serum Irisin decreased significantly (P< 0.05).(4) Serum level of Irisin were positively correlated with FAT%,BMI, TG, TC, FFA,2 HPBG, FIns,2 hins, FAI, HOMA-IR, H0MA2-IR (P< 0.05 or P< 0.01), and has negative correlation with the M value (P< 0.01).Multiple stepwise regression analysis showed that FAT % are independent risk factors that influence the level of serum Irisin. The higher the concentration of Irisin, the higher the risk of PCOS.Conclusion:Serum Irisin level is higher in Women with PCOS and associated with obesity and insulin resistance,it may be involved in the physiological and pathological process of obesity, insulin resistance and PCOS and may be a biological marker of obesity and insulin resistance. |
