OBJECTIVETo investigate whether the Galectin-7 exist in the extracellular of esophageal squamous cell carcinoma Eca-109 cells and the mechanism of Galectin-7 in the progress of esophageal squamous cell carcinoma。METHODS(1)Detecting whether the Galectin-7 exist in the extracellular of esophageal squamous cell carcinoma Eca-109 cells by means of Western blot。(2)Treating esophageal squamous cell carcinoma Eca-109 cells with human recombinant Galectin-7 of different concentrations; Treating esophageal squamous cell carcinoma Eca-109 cells with beta lactose。(3) Detecting the expressions of MMP-9 gene transcription by means of Real-time quantitative PCR; Detecting the expressions of MMP-9、p38 and pp38 protein by means of Western blot, Observing cell migration by means of scratch test; Observing cell multiplication by means of MTT test。RESULTS(1)The culture supernatant of Eca-109 cells can contain the Galectin-7。(2)The expressions of MMP-9 and pp38 were increased by human recombinant Galectin-7 and the ability of migrating of Eca-109 also had been enhanced,But the ability of multiplicating did not change obviously.(3)The expressions of MMP-9 decreased after treating with beta lactose。CONCLUSION(1)The Galectin-7 can be secreted to the extracellular by esophageal squamous cell carcinoma Eca-109 cells.(2)The Galectin-7 increased the expression of MMP-9 by activating p38MAPK pathway, And then affect the metastasis of Eca-109 cells.(3)The effect of Galectiri-7 can be restrained by beta lactose.
