ObjectiveThe present research,based on the locomotor activity in mice, the amino acid level in the cerebral cortex, as well as the distribution kinetics of ephedrine, pseudoephedrine in mice’s brain tissue,aimed to characterize the effect of Herbal ephedrae, Cinnamomi Ramulus and compatibility of the two herbs.The purpose of this paper is to find the relationship between the two herbs through detecting pharmacodynamic and pharmacokinetic effects on the central nervous system.Method1. Herbal ephedrae, Cinnamomi Ramulus were medicinal quality evaluation in accordance with the method of Chinses Pharmacopoeia. The concentration of ephedrine, pseudo-ephedrine in the water extract of Herbal ephedrae, compatibility of Cinnamomi Ramulus and Herbal ephedrae were detecting by GC-MS.2. The crossing and rearing counts of mice in the control group, the Herbal ephedrae group, the Cinnamomi Ramulus group and the combination group were recorded at 20 min,1 h,2 h,3 h, 4 h after medicine administration.3. With the use of high performance liquid chromatography, the 17 kinds of amino acids in the cerebral cortex were measured in the control group, the Herbal ephedrae group, the Cinnamomi Ramulus group and the combination group at 20 min,30 min,1 h,1.5 h,3 h and 5 h after medicine administration, in order to find difference content of each amino acid between the groups.4. Mice were ig adminstered by Cinnamomi Ramulus and codecoction of Cinnamomi Ramulus combined with Herbal ephedraee Herba. The content of ephedrine (E), pseudoephedrine (PE) in mice’s brain were determined by GC-MS. The non compartmental kinetic was fitted and the parameters were calculated by Kinetica 5.0.11 software, so as to evaluate the effect of Cinnamomi Ramulus on the distribution kinetics of E and PE from Herbal ephedrae in brain tissues of mice.Results1. The weight of Herbal ephedrae was counted on dry goods, including the total amount of ephedrine hydrochloride (C10H15NO·HCl) and pseudoephedrine hydrochloride (C10H15NO·HC1) to 1.42%(> 0.80%); The weight of Cinnamomi Ramulus was counted on dry goods, including cinnamic aldehyde (C9H8O) 6.42%(> 1.0%). The content of ephedrine, pseudoephedrine in Herbal ephedrae decoction were 0.52 mg/g,0.44 mg/g, the content of ephedrine, pseudoephedrine in codecoction of Cinnamomi Ramulus combined with Herbal ephedrae were 0.22 mg/g,0.20 mg/g.2. Compared with the control group, the mice in the Herbal ephedrae group significantly increased the level of activity counts at the time of 3 h after drug administration(P<0.05), there is a tendency to reduce the level of activity counts than mice compatibility Herbal ephedrae set of values.3. Compared with mice in control group, while 1.5h, Herbal ephedrae significantly reduced the level of inhibitory neurotransmitters amino acid (GABA, Ala), valine (Val), tyrosine (Tyr) (p O.05) in the brain of mice.4. Compared with Herbal ephedrae group, Herbal ephedrae-Cinnamomi Ramulus compatibility group delayed the time to peak(tmax), shortened mean residence time (MRT (0-t)), reduced the area under the curve (AUC (when drug 0-t)) of ephedrine, pseudoephedrine in brain tissues.Conclusion1. Herbal ephedrae has a central stimulant.2. Herbal ephedrae, Cinnamomi Ramulus and combination with two herbs can antagonize the impact on the concentration of amino acid, the amino acid neurotransmitters maintain this level with a steady lowering the side effects of Herbal ephedrae.3. Herbal ephedrae, Cinnamomi Ramulus compatibility compatibility can delay ephedrine, pseudoephedrine distribution process in the brain tissue of mice, reducing the amount of cumulative distribution in the brain, while accelerating its elimination in the brain.4. Cinnamomi Ramulus compatibility can reduce the excitability of the side effects of Herbal ephedrae.
