| Objective:The article aims to study the effect of electroacupuncture on spatial learning and memory, and on Ca2+ concentration and activity of CaM in related brain tissue. and to discuss the mechanism of electroacupuncture improving spatial learning and memory in morphine withdrawal rats and to provide credible experimental reference for accupuncture treating learning and memory deficit after morphine withdrawal.Method:56 male SD rats, of which weight between 160-220g, caged separately and fed for one week, then divided randomly into four groups:control group, model group, acupuncture group and electroacupuncture group. Rats in model groupã€acupuncture group and electroacupuncture group were injected with morphine subcutaneously within 5 days(20mgã€30mgã€40mgã€50mã€50mg), twice per day (8amã€4pm), rats in control group were injected with the same amount of saline at the same time. Naloxone (3mg/kg) was injected after the final dose of morphine, then rats were observed in beaker of 3000ml for scoring withdrawal symptoms. The one got higher scores can participate the next experiment.Four groups were treated separately after the model of morphine withdrawal rats were established, rats in acupuncture group were acupunctured Shentingã€Baihui, bilateral Shenshuã€Housanli for 6 days,15min per day after being tied. Rats in electroacupuncture group were acupunctured Shentingã€Baihui, bilateral Shenshuã€Housanli and connect the EA(2/100HZ,0.5-1-2mA,15min) for 6 days. Rats in control group and model group were tied at the same time. Rats were trained and tested for Morris water maze after acupuncture. Brain tissue were taken after the experiment to detect intracellular Ca2+ concentration and activity of CaM of different groups by flow cytometry. The data was analyzed by SPSS 17.0 Statistics, the final results were formulated in x±s, P<0.01 was of extremely significant difference, while P<0.05 was of significant difference.Results:(1) compared with control group, rats in model groupã€acupuncture group and electroacupuncture group all got higher scores, and the difference was believed to be statistically meaningful(P<0.01), which indicated that the model of morphine withdrawal rats were successfully established. (2) Morris water maze test:we compare each group and find out that electroacupuncture group and acupuncture group showed significant decrease in escape latency and increase in the frequency to swim across the platform (P< 0.01). (3) Compared with control group, Ca2+ concentration and activity of CaM showed a markedly decrease in model group(P< 0.01). In contrast to model group, Ca2+concentration and activity of CaM in acupuncture group and in electroacupuncture group increased markedly (P<0.01), and the difference between acupuncture group and electroacupuncture group was remarkable.Conclusion:These findings indicated:electroacupuncture can increase Ca2+ concentration and activity of CaM in which way to improve spatial learning and memory in morphine withdrawal rats. |
