Objective:To study the effects of morphine withdrawal rats for spatial learning-memory and the memory elements of calcium/calmodulin-dependent protein kinase Ⅱ (CaMK Ⅱ) activity in related brain areas by electroacupuncture, and try to explore the central nervous system mechanisms of EA improved the learning-memory in morphine withdrawal rats, provide experimental basis for the EA detoxification.Methods:We selected48SD male rats that weight ranging from160g to220g, feeded one week suitability. Rats were divided randomly into four groups, each group of12:control group, model group, electroacupuncture group, moxibustion group. In addition to the control group,the other groups were injected morphine hydrochloride injection by subcutaneous injection in back day by day,5days in all (twice a day).3h after the last injection rats were injected naloxone injection by intraperitoneal injection for rapid withdrawal. The model of rat morphine withdrawal established quickly.We observed different groups rats by different methods:(1)EA group rats were given a fixed bundle, acupuncture Baihui、Shenshu、Sanyinjiao Zusanli points, connect the EA, the density wave frequency2/100Hz,15min/once、once/d,treat6d;(2) Control group with the same time of the EA group fixed;(3) Model group with the same time of the EA group fixed;(4) Moxibustion group with the same time of the EA group to take the same acupuncture points and moxibustion treatments. Observed in the experiment (1) We observed the weight of rats in each group at different time points.(2) Each rat was mensurated by Morris water maze, navigation test to test spatial learning ability, space exploration experiments to test their spatial memory.(3) Immunohistochemical method was used to observe the changes of CaMK II in rats’VTA brain area.The SPSS17.0Statistics software was used to analyse datas of variance, P<0.05said significant differences, P<0.01said significant differences statistically.Results:(1) Compared with control group, the other groups all appeared the morphine withdrawal syndrome(P<0.01)for example:teeth chatter, wet dog shakes, diarrhea, lacrimation, salivation, jumping, weight loss etc. The model of rat morphine withdrawal established successfully.(2) After morphine withdrawal and treatment, compared with control group, there were significant differences in weight, such as reduce evidently or increase slowly in model groups (P<0.05). Electroacupuncture can promote weight gain in rats after withdrawal and restore the normal trend. Compared with control group, there were significant differences in it (P<0.05, P<0.01).(3) The results of Morris water maze indicated that compared with model group, the flat roof escape delitescence was cut down evidently by electroacupuncture (P<0.01), in the space exploration experiments, electroacupuncture and moxibustion can increase the number of across the platform (P<0.01).(4)The results of immunohistochemical method indicated that compared with model group, the expression of CaMK Ⅱ was heighten in VTA brain area of rats in electroacupuncture group of significantion differences (P<0.01)Conclusions:(1) EA can alleviate the symptoms of morphine withdrawal, and promote weight gain in rats after morphine withdrawal and restore the normal trend meanwhile alleviate withdrawal symptoms.(2) Electroacupuncture can shorten the flat roof escape delitescence of Morris water maze in order to improve learning ability of rats after withdrawal, and increase the number of pass platforms in order to improve memory ability.(3) Electroacupuncture can adjust the expression of CaMK Ⅱ, improve the active of CaMK Ⅱ to treat the learning-memory.
