Experimental Animal Study Of Delaying Renal Damage In Obstructive Nephropathy With Oral Aliskiren

Objective:To establish animal model of complete unilateral ureteral obstruction in New Zealand rabbits and study the effect of oral aliskiren delaying renal fibrosis and protecting renal function.Methods: The animal model of complete unilateral ureteral obstruction in healthy adult New Zealand rabbits was established by ligating the left upper utrter and peritoneal cavity.16 rabbits modeled successfully were randomized into test group and control group,and those in test group were given oral aliskiren 25mg/kg per day one week after operation until the test was ended.All rabbits in the two groups underwent renal ECT and Doppler ultrasound before operation and at 2nd,4th and 8th week after operation,and the specimens of the left kidney of all rabbits were collected for pathological examination when the test was ended.Result: The glomerular filtration rate(GFR) in the two groups were 28.4±1.7,27.4±1.8 respectively before operation,There was no significant statistically difference between the two groups.The GFR values of left kidneys of rabbits in test group were 11.2±1.5ml/min, 5.2±0.7ml/min, respectively at 2nd,4th and 8th week after operation,while that in control group were 6.3±0.5ml/min,2.8±0.2ml/min and 0.8±0.4ml/min respectively.The differences between test group and control group were statistically significant(P<0.05).Values of renal blood flow resisitant index(RI) of left kidney of the two groups were 0.60±0.03,0.59±0.01, There was no significant statistically difference between the two groups.Values of renal blood flow resistance index(RI) of left kidney in test group were 0.67±0.02,0.68±0.02 and 0.68±0.01 respectively at 2nd,4th and 8th week after operation,while RI values in control group were 0.68±0.03,0.70±0.02 and 0.75±0.02 respectively at 2nd,4th and 8th week after operation. Only at the 8th week The differences between test group and control group were statistically significant(P<0.05).Kidney slices HE staining shows that the degree of inflammation and fibrosis of left kidney in test group was weaker than that in control group at the eighth week after operation.Conclusions:It’s feasible and practicable to establish animal model of unilateral ureteral obstruction by ligating theutrter at ureteropelvic junction via peritoneal.Oralaliskiren is effective to delay renal fuction damage in obstructive nephronpathy through slowing down the renal inflammation reaction an fibrosis process.
