Objective: To detect the differential expression of ING4,VEGF and CD34 in the prostate cancer and benign prostatic hyperplasia,and investigate the relationship between the expression of ING4, VEGF, CD34 and pathological stage, risk rating to evaluate the role of ING4、VEGF、CD34 in progression of prostate cancer, as well as providing experimental basis for gene therapy of prostate cancer.Methods: To detect the expression of ING4、VEGF and CD34 in 28 prostate cancer samples and 32 benign prostatic hyperplasia samples from the Department of Pathology,Nanchong central hospital from Jan, 2012 to Dec, 2014 using immunohistochemical staining with ElivisionTM plus two-step method.Results:1. The positive rate of ING4 in benign prostatic hyperplasia and prostate cancer was 93.75% and 50% respectively(P<0.05). The positive rate of ING4 was 100% and 41.66% in middle differentiated and poorly differentiated prostate cancer respectively. The positive rates of ING4 was 100%, 85.71%, 23.53% respectively in PSA<10ng/ml, PSA 10-20ng/ml and PSA > 20ng/ml prostate cancer samples. The prostate cancer samples in clinical stage II, III and IV,the positive rates of ING4 was 100%, 76.92%, 8.33% respectively. There are significant differences between the positive expression of ING4 in prostate cancer samples of different pathological grading, clinical staging and PSA in prostate cancer rate(P< 0.05).2. The positive rate was 90.63% and 100%in benign prostatic hyperplasia and prostate cancer of VEGF, and there was no statistical significance between the two groups(P > 0.05). But the strong positive expression rate of VEGF in prostate cancer and benign prostatic hyperplasia was 60.71% and 31.25%respectively(P<0.05).The positive expression rate of VEGF in prostate cancer rose with tumor pathological grade, clinical stage and PSA increased(P < 0.05).3. The ratio of over 90 of micro vessel density(MVD)labeled by CD34 in benign prostatic hyperplasia and prostate cancer tissues was 31.25% and 78.57%(P< 0.05).The MVD increase with tumor pathological grade, clinical stage and PSA rose in prostate cancer tissues(P < 0.05).4. Spearman rank correlation was used to analyze the relationship among ING4, VEGF and MVD expression in the prostate cancer, results shown there is negative correlation between ING4 and VEGF, ING4 and MVD and VEGF was positively related to MVD.Conclusion:1. Low or lack of expression of ING4 in prostate cancer tissues indicates that ING4 can act as a tumor suppressor gene in the pathogenesis of prostate cancer and it may be used as a negative indicator of prognosis.2. The expression of VEGF and CD34 in prostate cancer tissues was significantly higher than that in benign prostatic hyperplasia tissues, and their expressions obviously correlated with the pathological stage and grade of prostate cancer,it suggest that high expression of VEGF may relate with the invasiveness of tumor growth.3. ING4, VEGF and CD34 may play important roles in the onset and progression of prostate cancer,indicating ING4, VEGF and CD34 can be used as valuable indicators for diagnosis and treatment in prostate cancer.
