Objects: investigating the expression of VEGF,COX-2 protein and MVD,nm23mRNA and their relations in esophageal carcinoma to provide theoretic basis and reference index for the diagnosis and treatment of esophageal carcinoma. Material and methods:1 In this study, 35 cases surgically resected esophageal carcinoma and 6 cases normal esophageal mucosa.2 The expression of VEGF protein and MVD(CD34 marked) were detected using immunohistochemistry staining.3 The expression of COX-2 protein was determined by Flow cytometry (FCM) in esophageal lesion.4 The expression of nm23mRNA was determined by in situ hybridization.5 Image Analysis :The results of immunohistochemistry and ISH are applied to HPIAS-1000 image analysis system and translated to gray values.6 Student't-test and linear correlation were carried out for statistics evaluation ,significance was set at P≤0.05.Result:1 The results of immunohistochemical staining1.1 The result of VEGF protein in esophageal carcinoma: there was a little VEGF protein in normal esophageal mucosa, the expression of VEGF protein in well differentiated, moderately differentiated, poorly differentiated carcinoma were higher and higher , there was statistic significance any two of them (P<0.05). The expression of VEGF protein in esophageal carcinoma were associated with their lymphatic nodes metastasis (P<0.05) but not with gender, sites, gross classification and infiltration degree (P>0.05). 1.2 MVD: the positive expression of CD34 in microvascular endothelial cell, the MVD of normal esophageal mucosa was lower than carcinoma tissue. There was associated with their lymphatic nodes metastasis (P<0.05) but not with age, gender, sites, gross classification and infiltration degree (P>0.05) .2 Analysis result of FCMThe FI of COX-2 protein in normal esophageal mucosa was lower than its expression in esophageal squamous cell carcinoma (p<0.05).The FI of COX-2 protein in well, moderately and poorly differentiated was higher one by one, there was statistic significance between both of them(P<0.05).The expression of COX-2 protein was associated withtheir lymphatic nodes metastasis but not with gender, sites, gross classification, infiltration degree (P>0.05).3 The result of nm23mRNA in situ hybridization Nm23mRNA expressed in every normal mucosa, cancer tissue, there was statistic significance between both of them(P<0.05) , the expression of nm23mRNA was associated with their lymphatic nodes metastasis but not with gender, sites, gross classification, infiltration degree, grade of dysplasia and grade of differention (P>0.05).4 The relation between VEGF, MVD, COX-2 and nm23mRNA expression There was positive correlation between VEGF and MVD, VEGF and COX-2, MVD and COX-2, one of then increased with the other's increasing in esophageal normal mucosa, squamous cell carcinoma. There was no correlation between nm23mRNA and any other of them.Conclusions:1 The expression of VEGF and COX-2 protein in normal esophageal mucosa ,the side of carcinoma and squamous cell carcinoma became higher one by one ,its showing that the VEGF and COX-2 protein expression increased in esophageal squamous cell carcinogenesis. The result suggested that VEGF and COX-2 maybe the index of the esophageal mucosa's carcinogenesis, poorly differentiated, lymphatic node metastasis and vessel invasion. 2 CD34 marked MVD increased in normal esophagealmucosa, dysplasia tissue and squamous cell carcinoma, suggest that the MVD was associated with esophageal squamous cell carcinogenesis, poorly differentiated lymphatic node metastasis and vessel invasion.3 The expression of nm23mRNA is decreased in normal esophageal mucosa dysplasia tissue and squamous cell carcinoma, its suggested that the nm23mRNA was associated with esophageal squamous cell carcinogenesis, and the expression of nm23mRNA was associated with lymphatic node metastasis, suggested that the high expression nm23mRNA was an index of esophageal squamous cell carcinoma's...
