Tripolide,a kind of Epoxy two compound extracted from Chinese herbal plant Tripterygium wilfordii,is a main active component as well as main toxic component. In both clinical and experimental studies, It has been proved that the extract from the plant is able to prolong the growing season of the graft and prevent the occurrence of graft versus host disease effectively. However, these extracts including TP in the regulation mechanism of CD4+CD25+ regulatory T cells in the immune suppression has not been fully studied. With the deeper studies of triptolide, its drug effective value has been widely acknowledged. In this study, we will research on its mechanism of anti immune rejection as well as provide some theorical basis for the treatment of immunological rejection after penetrating corneal transplantation.Through the single factor experiment and the establishment of penetrating corneal transplantation on Wistar rats model, we analyzed the CD4+CD25+ relugary cells and related cytokines in the rats of the same kind of corneal transplantation. The 48 Wistar rats were divided into two groups with 24 rats in each group, 16 for the recipient and 8 for the donor. Two groups were respectively, the rats in the blank control group(A group), and the rats injected triptolide(B group). We used the cornea to establish an animal model of Wistar rats through replacement allograft. 7 days, 10 days, 14 days after operation, data were collected by cardiac blood sampling method to study enzyme linked immunosorbent assay in both A and B Group. On the fourteenth day after surgery, we collect the related data on peripheral blood, lymph nodes and spleen of A, B two groups of experimental animals were collected by FCM.According to the observation of cornea and the evaluation of various indexes, we found that the degree of edema, opacity of graft and the entering of new vessels in experimental group are lower than the control group. Detection of peripheral blood in rats at the 7th, 10 th, 14 th days showed that the levels of IL-10 in the experimental group were significantly increased especially on the 14 th day. Contrary to the experimental expectation, the level of TGF-β 1 was markedly decreased. The results of flow cytometry showed that the CD4+CD25+n Treg level of the experimental group was significantly higher than the control group in the peripheral blood, spleen and lymph nodes in the 14 day after operation.In summary, triptolide intraperitoneal injection for ten days before and after the operation can increase the amount of CD4+CD25+ n Treg and IL-10 and suppress the secretion of TGF-β1.
