| Objective:To explore the differentiation of bone marrow mesenchymal stem cells (MSCs) indirectly co-cultured with purified cardiomyocytes (CMs) of neonate rats. Methods:Third generation of MSCs were indirect co-cultured with CMs through transwell in a rate of 1:5 for 3 days and 7 days, respectively, and MSCs cultured without neonatal CMs served as controls. CMs marker including a-actuu myocardial troponin T (cTnT) and myocardial troponin I (cTnl) were determined by immunofluorescence staining on day 3 and day 7.Results:Positive immunofluorescence against α-actinã€cTnT and cTnI were detected in MSCs co-cultured with CMs but not in MSCs cultured without CMs.Conclusion:Indirect cell-to-cell contact between MSCs and CMs may induce MSCs differentiation into cardiomyocytes-like cells. |
