Objective: To investigate the effect of concurrent chemo-radiother apy with temozolomide on glioblastoma multiforma(GBM).Methods: The GBM patients in the first hospital of Jilin university and Tumor hospital of Jilin province were recruited from 2009.01 to 2012.12. Then the patients were grouped according to the wills into radiotherapy alone group and radiotherapy plus temozolomide group. The radiotherapy alone group were given l irradiation. The radiotherapy plus temozolomide group were given radiotherapy and temozolomide(75 mg/m2/d, 7d/a week from the first to the last day of radiotherapy, no more than 49d) + 4 weeks interval + adjuvant temozolomide(150 to 200 mg/m2 for 5 days during each 28-day cycle). The KPS scores, two-years survival rate, the progression-free survival time, safety analysis and the quality of life were compared between the two groups.Results: 1. The two-year survival rate in radiotherapy plus temozolomide group was higher than radiotherapy alone group(P=0.0345). The median two-year survival time was 61 weeks with radiotherapy plus temozolomide group and 52 weeks with radiotherapy alone group.2. The two-year progression-free survival rate in radiotherapy plus temozolomide group was higher than radiotherapy alone group(P=0.047). The median two-year progression-free survival time was 29 weeks with radiotherapy plus temozolomide group and 21 weeks with radiotherapy alone group.3. Concomitant treatment with radiotherapy plus temozolomide resulted in similar side effcets compared to radiotherapy alone(P>0.05). The frequencies of grade 3 or 4 hematologic toxic effects and severe fatigue were higher in concomitant treatment froup(P<0.05), however, the toxic effects quickly recovered after 1-2 weeks.4. The concomitant treatment with radiotherapy plus temozolomide group inproved the quality of life of patients with glioblastoma multiforma(P<0.001).Conclusion: The concurrent chemo-radiotherpy with temozolomide on glioblastoma multiforma resulted in significant survival benefit with minimal side effects.
