Objective:By observing the Yupingfeng particle intervention model rats with allergic rhinitis in peripheral blood of CD19, CD23 expression of lymphocytes and the influence of nasal mucous tissue pathology change,to research the mechanism of Yupingfeng particle treatment of allergic rhinitis and provide experimental basis for the clinical application.Methods:66 SD rats were randomly divided into normal control group of 20, model group of 40, passive smoking device using self-designed experimental rats by the method of smoke lung deficiency of syndrome animal model is set up, in the rat trachea injection of Lipopolysaccharide(LPS)200μg/200μL, twice a day in the red elaborations of cigarettes smoked in the 16 and a half hours. On this basis, in addition to the normal control group,each rat with Ovalbumin(OVA), on the basis of sensitization and local two phase of Allergic rhinitis animal model, AR model group were randomly divided into model group and Yupingfeng particles groups, each group of 20. After the success of the building, the Yupingfeng particles to Yupingfeng particles group rats’ lavage, 1 times a day, 7 days of continuous larvae. Normal control group and the group with normal saline instead of AR model. The second day after the treatment, 1 ml venous blood collection, CD19 in peripheral blood of rat, CD23 and CD19/CD23 tested lymphocyte percentage analysis;Each group to collect other 1 ml venous blood, take the serum, centrifugal after the IgE-ELISA kit to detect serum total IgE expression. Then take the nasal mucous membrane of the breathing zone, by immunohistochemistry technique to detect the CD23 content in nasal mucous of rats. All statistics using SPSS17.0 statistical software for processing,groups of calculated result is expressed the average ± standard deviation( x ±S), statistical methods using single factor analysis of variance between groups compared, f test, before comparing to P < 0.05 for the difference was statistically significant.Results:1. Semeiology score: normal control group score for(1.69±0.21) <5 points, AR model group score(7.82±0.63)>5 points, Yupingfeng particles group score(2.85±0.31)<5.2. Nasal secretion smears observation: normal control group after treatment,respectively, nasal secretion smear, then not seen in nasal secretion smear eosinophils and mast cells. AR model group, then the vast majority of nasal secretion smears of visible to the eosinophils and mast cells. Yupingfeng particles group visible part seen in nasal secretion smear to eosinophils and mast cells.3. The rat serum total IgE content: AR model group compared with normal control group, serum total IgE content increased obviously, the difference was statistically significant(P<0.05); Yupingfeng particle group compared with AR model group, the serum total IgE content decreased significantly, the difference was statistically significant(P<0.05); Yupingfeng particle group compared with normal control group, serum total IgE content close no statistically significant difference(P>0.05).4.The rat peripheral blood of CD19,CD23 and CD19/CD23 expression of lymphocyte percentage: AR model group compared with normal control group during the period of onset has increased trend, the difference was statistically significant(P<0.05), Yupingfeng particle group compared with AR model group all has a tendency to reduce after drug intervention, the difference was statistically significant(P<0.05), Yupingfeng particle group compared with normal control group by statistical analysis, the difference is not significant(P>0.05).5. The expression of CD23 content in nasal mucous of rats: epithelium goblet cells found that AR model group was obviously increased, sub-mucous tissue edema,inflammatory cells infiltration and the increase in the number of eosinophils gland hyperglycemia, small blood vessels increase, congestion, increase in the number of CD23 marked cells specificity; Yupingfeng particles group organization slightly edema, tiny blood vessels, glands local hyperglycemia and inflammatory cells infiltration degree of improvement.Conclusion:1. Immunohistochemistry and flow cytometry analysis results confirmed that the Yupingfeng particles can regulate the expression of CD19 and CD23 in B lymphocyte.2. Yupingfeng particles can relieve nasal tissue inflammation, delay the pathological process.
