Experimental Study On The Choosing Of Acupoints "Jiangzhi Decoction" And On Abnormal Blood Lipid In Rats Of The Lipid Regulating Effect

Objective To screen the best prescription of acupoints with comparing the clinical regulate dyslipidemia based on “Quchi, Zhongwan, Fenglong ” of the most commonly used acupoints of different combinations of effect.Based on those results, observe the effect of the best point combinations on ET-1, abnormal blood lipid in rats NO, Ox-LDL content, and to explore its possible mechanism of regulating effect on blood lipid.MethodExperiment one One-hundred and six male SD rats were randomly divided into 9 groups,including a normal group, a model group, a Quchi group, a Zhongwan group, a Fenglong group, a Quchi+Zhongwan group, a Quchi+Fenglong group, a Zhongwan+Fenglong group, a Quchi+Zhongwan+Fenglong group, 18 rats in the normal group and 11 rats in the rest group. the normal group fed with basic feed every day, daily ration to high fat diet for other groups, drinking water is unlimited, continuously feed freely for 7 weeks. The 3 weekend in the blank control group, 8 rats randomly selected from 1 other groups, content selection, abdominal aortic blood and the detection of TG, TC, LDL-C and HDL-C, observe model was successfully established. Fourth weeks of starting treatment, and draw material after 4-weeks. Detecting the content of rats in each group TC, TG, LDL-C, HDL-C, ALT and AST, observed the changes of liver, aorta and fat cells under the light microscope.Experiment two thirty male SD rats were randomly divided into 3 groups, including a normal group, a model group, a “Jiangzhi Decoctionten” group, rats in each group, Reference experiment one for modeling and acupuncture. For 7 week sampling, detecting the content of ET-1, NO and Ox-LDL in rats of each group.Result1: At the end of three weeks of modeling, compared with normal group, content of rat serum TC, TG and LDL-C increased significantly(P < 0.05), decrease the content of HDL-C(P < 0.01) in the model group.Which shows that the model rats were successfully prepared with dyslipidemia. After the treatment, the contents of TG and LDL-C in TC model group, is still high(P < 0.01), decrease the content of HDL-C(P < 0.01), it’s indicated that the rats has been in a state of abnormal blood lipids in the experimental period.2. Compared with the model group, rats serum TC, TG, LDL-C, HDL-C, ALT and AST contents were significantly decreased(P < 0.05, P < 0.01)in the seven EA group, "Quchi + Zhongwan + Fenglong" group and EA group compared with the other 6, The TC, TG, LDL-C, ALT and AST decreased significantly, and the content of HDL-C increased significantly, And the values tend to normal group.3. Under the light microscope: the model group rats liver visible cytoplasm gathered a large number of lipid droplets, fatty degeneration, cell of moderate to severe swelling, part of the cell membrane disappeared, mutual integration, HE staining showed the size of the vacuole, disrupting the normal structure of hepatic lobules; thoracic aorta endothelial cells and muscle layer no abnormal lesions, indicating no occurrence of early AS lesions; fat cell volume increases, reducing the number of view within the same cell, the fat cells cytoplasm rupture, mutual integration, and the formation of vacuoles became larger than before. The liver cells, fat cells in each electroacupuncture group which has different levels of repair, and "Quchi + Zhongwan + Fenglong" group improved more obviously.4. Compared with the normal group, increase the content of ET-1, Ox-LDL of the rats in the model group, NO decreased significantly(P < 0.05),the "Jiangzhi Decoction" group has no difference on ET-1, Ox-LDL and NO(P>0.05). After acupuncture, "Jiangzhi Decoction" group compared with model group, ET-1, Ox-LDL content decreased, NO increased significantly(P < 0.05).Conclusion1.Electroacupuncture at "Quchi + Zhongwan + Fenglong acupoint" group reduced optimal effect of blood fat, so it can be used as the basic prescription of acupoints regulating blood lipid abnormality.2.Effects of Electroacupuncture of "Jiangzhi Decoction" acupoint prescription can effectively reduce the ET-1, Ox-LDL level, increase the level of NO, it is found that acupuncture is the possible mechanism regulating blood lipid abnormality because its efficacious for improving endothelial function and antioxidant capacity.
