Objective: To study the chemical constituents, the antioxidant activity and the totalflavonoids of Rhamnus songorica’s branches, leaves and fruits preliminarily. To observe thetherapeutic effects of Rhamnus songorica Extract (RSE) of fruits in rats with regulatingblood lipid and antioxidant by means of establishing the rat model of hyperlipidemia.Methods: The methods of systematic pretest and individual pretest were adopted to studythe chemical constituents of Rhamnus songorica’s branches, leaves and fruits. Todetermine the total flavonoids by spectrophotometry and DPPH,·OH, O2-· methods wereconducted to evaluate the antioxidant activity of different medicinal parts in vitropreliminarily. A rat model of hyperlipidemia was established. Positive control group wasgiven zhibituo (187.5mg/kg). The low-dose RSE group, middle-dose RSE group andhigh-dose RSE group were given RSE75,150,300mg/kg respectively. The levels ofbiochemical parameters related in serum, hepatic tissue and the evaluation of thehistopathological changes of hepatic and aortic on rats were detected after4weekstreatment. Results: The branches of systematic pretest on carbohydrates, tannins, saponins,quinoids, coumarin and macrolides, organic acids, flavonoids, phenols, oil fat was positive,and the individual pretest on anthraquinones was positive. The leaves of systematic preteston amino acids, polypeptide, proteins, carbohydrates, tannins, organic acids, flavonoids,phenols was positive. The fruits of systematic pretest on carbohydrates, tannins, quinoids,organic acids, flavonoids, phenols was positive, and the individual pretest onanthraquinones was positive. The result of spectrophotometry was that the total flavonoidscontents in branches, leaves and fruits of RSE were1.10%,2.42%and11.41%respectively. The antioxidant activity on DPPH,·OH and O2-· in vitro was fruits>leaves>branches. The therapeutic effects of RSE in rats with hyperlipidemia:compared with the rats in normal group, the level of TC, TG, LDL-C, ALT, AST, MDA,NO significantly increased (P<0.01), the level of HDL-C, T-SOD, GSH-PX, HL, LPL, LAsignificantly decreased (P<0.01) and hepatocyte, aortic steatosis were observed in ratswith hyperlipidemia. Compared with the rats in model group, RSE significantly decreasedthe level of TC, TG, LDL-C, ALT, AST, MDA, NO and obviously increased the HDL-C,T-SOD, GSH-PX, HL, LPL, LA content after4weeks treatment with RSE. The hepaticand aortic pathological changes were milder in the RSE groups than in the model group.Conclusion: Rhamnus songorica branches contain carbohydrates, tannins, saponins,quinoids, coumarin and macrolides, organic acids, flavonoids, phenols, oil fat. Rhamnussongorica leaves contain amino acids, polypeptide, proteins, carbohydrates, tannins,organic acids, flavonoids, phenols. Rhamnus songorica fruits contain carbohydrates,tannins, quinoids, organic acids, flavonoids, phenols. The total flavonoid contents andantioxidant activity were fruits>leaves>branches. RSE of fruits can regulate blood lipid,increase the antioxidant capacity.
