The first part Eastablish the model of rats cerebral ischemia-reperfusion (CI/RP) after Maixuekang and aspirin pretreating and investigate the affect of the score of neurological function deficit and cerebral infarct volumesObjectives:To eastablish the model of rats CI/RP injury and investigate the effects of Maixuekang and aspirin pretreating in rats focal CI/RP injury through the score of neurological function defict and cerebral infarct volumes.Methods:40 male Sprague-Dawley(SD) rats(SPF,weight between 280-300g) were randomly divided into five groups:1.sham operation group (saline 4ml gavage 14 days),2.model control group (saline 4ml gavage 14 days),3.aspirin pretreating group (aspirin 10mg/(kg-d) gavage 14 days),4.Maixuekang pretreating group (Maixuekang 1.0g/(kg-d) gavage 14 days),5.the joint administration of pretreating group (Maixuekang 1.Og/(kg-d)+aspirin 10mg/(kg-d) gavage 14 days). After 14 days, the rats middle cerebral artery occlusion (MCAO) were performed to establish focal cerebral ischemia-reperfusion injury model. After 2h,6h,and 24h, neurological function deficit was scored.24h after the cerebral ischemia-reperfusion injury, the cerebral infarct volumes were calculated by TTC stain.Results:Suture method can establish a stable rats ischemia-reperfusion injury model. 6 and 24hours after ischemia-reperfusion,the neurological deficit scores of drug pretreating groups had significant differenenc with model control group.After 24 hours the cerebral infarct volumes had significant differenenc between drug pretreating groups and model control group. And the combined pretreating group had significant difference with Aspirin or Maixuekang pretreating alone group.Conclusion:Maixuekang or aspirin pretreating can reduce the CI/RP injury.the two drugs pretreating together have a better effect.The second part The affects of coagulation function and blood rheology after the CI/RP injury rats with Maixuekang and Aspirin pretreatingObjective:Examine the blood coagulation function and blood rheology to exploring whether Maixuekang and Aspirin reduce the CI/RP injury by improving coagulation function and blood rheology.Methods:The animal grouped and preteated as the first part,each group also had 8 rats. Based on the successfully establishing the ischemia-reperfusion injury model. 24 hours after the injury, the rats blood was obtained from abdominal aortic. the blood coagulation function and blood rheology were examined.Results:The coagulation fuction and blood rheology of the model control group had significant difference compared with the sham operated group (P<0.01). Drug pretreating groups had significant difference (P<0.05) with model control group.the two drugs pretreating together group had significant difference compared with the Maixuekang or Aspirin pretreating alone group (P<0.05).Conclusion:After CI/RP injury,the rats blood in hypercoagulable state and flowing slowly.Maixuekang and aspirin can change the hypercoagulable state and improve the blood rheology, the two drugs pretreating together has better outcomes.The third part The affects of Bcl-2, Bax, Caspase-3 in the Apoptotic pathway after CI/RP with Maixuekang and Asprin pretreating ratsObjective:To investigate the change of Bcl-2, Bax, caspase-3 protein expression in apoptosis pathway after CI/RP with Maixuekang and Asprin pretreating rats.Method:The animal grouped and preteating as the first part,4 rats each group.The rats were decapitated 24 hours after the CI/RP injury and obtained the brain tissue. Protein was extracted from infarcted area. Western bloting method was used to detect the expression of the B-cell lymphoma/leukemia 2 gene protein(Bcl-2), Bcl-2 associated X (Bax) protein and cysteinyl aspartate specific protein-3(caspase-3) protein.Results:Compared with the sham operated group, the expression of Bax, Caspase-3 protein increased and Bcl-2 protein decreased in model control group. Compared with the model control group, the expression of Bax and Caspase-3 protein decreased and Bcl-2 protein increased in drug pretreating groups. Compared with the aspirin group or Maixuekang group, the expression of Bax, Caspase-3 protein decreased and Bcl-2 protein increased in the twe drugs pretreating together group.The differences had significant difference.Conclusion:The protect effect of Maixuekang and aspirin pretreating rats after CI/RP injury may through up-regulating the expression of Bcl-2 protein and down-regulating Bax and Caspase-3 protein. Inhibited the cell apoptosis and protect brain cells. This protective effect is significantly better in the two drugs pretreating together group.
