Objective:Intraperitoneal injection of gel sensitization agent to establish rat intestinal dysbacteriosis merger model of allergic asthma, testing rats serum and alveolar lavage fluid (BALF) IL-10, IL-17, IL-35 cytokines, through the relevant regulation of Th17 and Treg cells factor content determination to explore Gui Zhi Jia Hou Pu Xing Zi Tang on intestinal dysbacteriosis combined allergic asthma rats curative effect and the effect of Th17/Treg balance,and provide experimental basis for the clinical use of Gui Zhi Jia Hou Pu Xing Zi Tang.Method:60 rats were divided into model group, blank control group, model group is divided into Gui Zhi Jia Hou Pu Xing Zi Tang high, medium and low dose group, positive control group, model control group. Intraperitoneal injection of model group 1 and 8 days to gel sensitization agent 1 ml,2-6 days feed cephalosporins pp sodium sodium shu ba tan drinking water, to drink the tap water instead of 7 days, at the same time to candida albicans 1 ml lavage,15 days to 21 days with 2% ovalbumin solution atomization 30 min/time/day inspire rats of asthma, blank group to drinking water, tap water to replace antibiotics to saline sensitization and atomization inhalation, after spray irrigation Gui Zhi Jia Hou Pu Xing Zi Tang high dose, medium, low dosage, aminophylline, irrigation model control group take physiological saline, blank control group without treatment, continuous lavage for 7 days,21 days pulmonary function testing,22 days by intraperitoneal injection of 10% chloral hydrate anesthesia in rats after materials, heart blood related blood smear, BALF smear, after gut bacteria count, serum and BALF of IL-10, IL-17, IL-35 cytokines index check, lung, intestinal tissue pathological biopsy.Results:Gui Zhi Jia Hou Pu Xing Zi Tang high, medium and low dose group and aminophylline group rats of asthma symptoms and signs improved; Lung function and intestinal flora better; Each treatment group EOS, IL-10, IL-35, IL-17 compared with model group, the difference were significant statistical significance.1.the content of the IL-10 in serum and BALF:Gui Zhi Jia Hou Pu Xing Zi Tang dose group was significantly higher than in other treatment group (P<0.01); Low dose group is higher than aminophylline group (P<0.05), the difference was statistically significant; No difference between the high dose group and aminophylline group, there was no statistically significant difference between high and low dose group.2.IL-35 content in serum and BALF:Gui Zhi Jia Hou Pu Xing Zi Tang dose group was significantly higher than in other treatment group (P<0.01); Serum low dose group was significantly higher than that of high dose group, aminophylline group (P< 0.01), high dose group is higher than aminophylline group (P<0.05); BALF high and low dose group is higher than aminophylline group (P<0.05), the difference was statistically significant, difference between high and low dose groups has no statistical significance.3.IL-17 content in serum and BALF:Gui Zhi Jia Hou Pu Xing Zi Tang was significantly lower than in other treatment group (P<.01); High and low dose group was significantly lower than that of aminophylline group (P<0.01), the difference was statistically significant. Differences between high and low dose groups has no statistical significance.Conclusion:Gui Zhi Jia Hou Pu Xing Zi Tang can lower intestinal dysbacteriosis merger Th17 allergic asthma rats secretion of IL-17 cytokines, increased Treg cell secretion of IL-10, IL-35 cytokines, show that the treatment of intestinal dysbacteriosis combined allergic asthma role may be balanced by influence of Th17/Treg cells related regulatory factor.
