Aim:Study on the treatment effect of Moxibustion and the influence of EA on IL-2 and IL-17A with Ulcerative Colitis(UC) mice model induced by DSS, and analysis the effect of plasma metabolite of Moxibustion therapy to UC mice model from the aspect of metabonomics. Preliminary exploration the metabolic mechanism of Moxibustion treatment on UC mice, to provide experimental basis for clinical acupuncture and moxibustion treatment of UC.Methods:24 SPF male KM mice continuous drinking 3% DSS to induce mice UC diseases, the mice were randomly divided into UC model group and moxibustion treatment group after successfully setup models, another 10 age weight of mice as control group. The treatment group choose "Zu San Li, Guan yuan" points treated with moxibustion from the fifth day after making model. Recording DAI, observed colon tissue morphology by optical microscope and transmission electron microscope, ELISA assay the content of IL-2、IL-17A in mice plasma of each group.HPLC-MS analysis the differences of blood metabolites, and compare effect of plasma metabolites of UC mice treatment with moxibustion.Results:1. The general curative effect observation:(1) In general:Control mice were in good spirits, hair light, normal stool. After model setup, mice were gradually appear lazy little food, hair color is dim, weight loss and fecal occult blood positive. After treatment spirit improvement, slightly shiny hair, stool soft, occult blood (weak) positive or disappear.(2) DAI score:After modeling, the model group DAI score increased. After treatment, moxibustion group DAI index decreased, the difference was statistically significant compared with the model group (P<0.05).(3) Colon tissue morphology:Under the light microscope, colonic mucosal tissue of model group structural incomplete, glands disappeared, goblet cells increased, ulcers and necrosis formation, a number of inflammatory cell infiltration; moxibustion treatment group part of gland destruction, mucosa mild edema, a few inflammatory cells infiltration. Under electron microscope, model group microvilli are few and short,vacuoles in cells, mitochondrial swelling;after the treatment, micro pile is more complete, mitochondrial swelling ease;control group were no pathological changes under the light microscope and electron microscope.2. Immune effect:model group compared with the control group IL-2 content significantly decreased while IL-17A content rise in blood after modeling, the differences were statistically significant (P<0.05). Moxibustion group compared with the model group, the IL-2 was significantly increased, but IL-17A is reduced, the differences were statistically significant (P<0.05) after treatment.3. Effects of plasma metabolites:Diacylglycerol carnitine, fluorouracil, butyrylcholinesterase, uric acid is the main metabolite difference. These metabolite associated with fatty acid, amino acid metabolism and energy metabolism pathway.Conclusions:Moxibustion is effective to cure UC mice, could improve immune cytokines IL-2, IL-17A and the diacylglycerol carnitine, fluorouracil, butyrylcholinesterase, uric acid etc metabolism disorder in UC mice plasma, prompt that Moxibustion maybe influence energy metabolism pathway which associated with these metabolism.
