Objective:To investigate the effects of the asarum、the aconite and the serum containing asarum and aconite on L-type calcium channel currents in isolated rat ventricular myocytes, study the mechanisms of antiarrhymias.Methods:Aconite and asarum decoction,compatiblility of water bath concentration 1:1, rats were given intragastrically, every time 2. 5g·kg-1,7d after preparation containing serum; obtained using enzymatic hydrolysis acute rat ventricular myocytes, current using whole-cell patch clamp technique join Asarum, aconite and compatibility of ventricular myocytes serum containing L-type calcium channels, current signal by the patch clamp amplifier, filtering, Pclmp 10.0 software acquisition by the current changes, clampfit software for data processing.Experimental program designed according to ventricular activation characteristis of L-type calcium channels, inactivation and recovery stimulus package.Current drawing activation, deactivation and recovery curve based on the record.Analysis Asarum, aconite and its prescription drug-containing serum on ventricular L-type calcium channels.Result:(1) The peak current display:the average activation fo blank control group、 asarum、aconite and compatibility groups respectively(12.87±0.25)%(n=8,p<0.05)、 (28.36±0.37)%(n=8,p<0.05)、51.17±0.25)%(n=8, p<0.01)�'� (75.07±0.28)% (n=8,P<0.01); (2) The current voltage (Ⅰ-Ⅴ) curve display:blank、asarum、 aconite and the compatibility group can depress the Ⅰ-Ⅴ curve, the compatibility of aconite and asarum group stronger activion peak current; (3)The steady-state activation curve display:with asarum、 aconite and the compatibility group to join, the steady-state activation curve shifted to left, to the opening of calcium channel plays a significant role in promoting; (4) The state inactivation curve display:with asarum、aconite and the compatibility group to join, the steady-state inactivation curve was shifted to the left, the acceleration of L-type calcium channel inactivation; (5) The steady-state inactivation curve after recovery:with asarum group and aconite group to join, the steady-state inactivation curve of recovery after no obvious effect, but with the addition of asarum compatibility of radix aconti group, the steady-state inactivation recovery time。Conclusion:(1) Asarum medicated serum, the activation curve of the inactivation curve to the left、 had no obvious effect on the recovery curve; aconite containing serum, the activation curve of the inactivation curve to the left、 had no obvious effect on the recovery curve; the serum containing asarum compatibility of radix aconiti the activation curve、the inactivation curve to the left, to shorten recovery time; (2) Activation of Asarum compatibility aconite containing serum on ventricular myocytes L-type calcium channel is stronger than the unit Asarum, aconite, show the compatibility of the two drugs have similar effects to be synergistic.
