Objective:To observation the Effect of enriched environment intervention on BDNF expression and apoptosis in hippocampus CA1 region of hypoxic-ischemic brain damage neonatal rats. To explore the mechanism of enriched environment on hypoxic-ischemic brain damage.Methods:7-day-old rats were randomly divided into sham operation group, model group, enriched environment group, each group was further divided into14 days and 28 days subgroups(n=6). HIBD model of neonatal rats was established according to Rice-Vannucci classical methods. The model group and the sham operation group accepted no treatment, enriched environment group was treated by enriched environment stimulation.14days,28days after operation, the learning and memory ability was evaluated with the water maze detection method, the positive expression levels of BDNF protein were detected by Immunohistochemical method. The apoptosis were detected by TUNEL method.Results:1. Learning and memory changes of rats in each group: Postoperative 14d and 28d, compared with the sham operation group, the model group the average escape latency period(EL) was significantly more prolonged, thecross-platform number within 2 minutes was significantly less, and the difference was statistically significant(P<0.05); Compared with the model group, the enriched environment group the average escape latency period(EL) was shortened, thecross-platform number within 2 minutes was increased, and the difference was statistically significant(P<0.05).2. Changes of the positive expression of BDNF protein in rats of each group:Postoperative 14d and 28d, compared with the sham operation group, the model group rats hippocampal CA1 visible positive expression of BDNF protein increased, the difference is statistically significant (P<0.05); compared with the model group, the enriched environment group in the CA1 region of the hippocampus BDNF protein positive expression was significantly, The difference was statistically significant (P<0.05).3. The positive changes in rats apoptosis:Postoperative 14d and 28d, compared with the sham operation group, the model group the positive expression of apoptosis increased, the difference is statistically significant (P<0.05); compared with the model group, the enriched environment group apoptosis decreased significantly. The difference was statistically significant (P<0.05).Conclusion:1. Enriched environment can improve the learning and memory ability of HIBD rats.2. Enriched environment can up regulate the expression of BDNF, and down regulate the positive expression of cell apoptosis in hippocampal CA1 area of HIBD rats.
