ObjectiveDiscussion on learning and memory ability and TrkB、p-Akt protein expression of Hippocampal CA1 region in hypoxic-ischemic brain injuried rats by enriched environment, providing theoretical support in clinical rehabilitation.MethodsA total of 367-day-old Sprague-Dawley rats were randomly divided into sham, HIBD, and environment enriched groups (n=12 each). The HIBD model was established using the modified Rice method,environment enriched are intervented by environment,sham-operated and HIBD are without intervention. afer established model 14 days and 28 days,choose the objects, The learning and memory ability was tested by Morris water maze, In hippocampus CA1 region of rats pathological changes was detected by hematoxylin-eosin staining (HE staining),The expression of TrkB、p-Akt protein in area CA1 of the hippocampus was measured by Western Blot.Results1. on learning and memory ability:Rat brains at 14、28 days after HI,compared with the sham groups, the average time of escape latency was significantly longer and the number of platform location crossing was more (P<0.05) in HIBD model groups.2. Histopathological changes:Rat brains at 14、28 days after HI, No histopathological changes in the area CA1 of the hippocampus of the sham-operated rats, Neural structure and morphology of the cells showed normal; cells edema were more in the area CA1 of the hippocampus of the model HIBD rats,loose tissue and vacuolar changes were more obvious and hypoxic-ischemic central nerve cells exhibit the characteristics of cell necrosis, inflammatory cell infiltration occurred. compared with the model HIBD groups,pathological changes in the area CA1 of the hippocampus of the environment enriched groups exhibited varying degrees of ease. the number of degeneration, necrosis was significantly reduced.3. the expression of protein TrkB、p-Akt:the stable expression of protein TrkB、p-Akt in the area CA1 of the hippocampus of the sham rats. Rat brains at 14、28 days after HI,compared with the sham,the expression of protein TrkB、p-Akt in the area CA1 of the hippocampus of the model HIBD rats was remAktly more(P<0.01). the expression of protein TrkB、p-Akt in the area CA1 of the hippocampus of the environment enriched groups is more than the model groups.Conclusion1. Enriched environment improves the ability of learning and memory in HIBD rats and reduces the pathological damage in hippocampal CA1 region, having neuroprotective effect.2. Enriched environment can upregulate the expression of protein TrkB、 p-Akt in the area CA1 of the hippocampus and effectively reduce hypoxic-ischemic brain injury.
