| Objective:Trapa manshurica Flerow was a natural medicine-food plant. Contains various of effective composition, such as polyphenols, polysaccharides, flavonoids and sterols. Trapa manshurica Flerow has many kinds of efficacy, such as anti-tumor, antioxidant and antibacterial. In order to more effectively to development and utilization of Trapa manshurica Flerow, the extraction technology and antioxidant activity were studied. Method:1. Study on extraction and antioxidants ability of polysaccharide in Trapa manshurica Flerow.Optimization for water extract-alcohol precipitation method of polysaccharide in Trapa manshurica Flerow by single factor experiment. Select material liquid ratio, extraction temperature and extraction time to study. Calculate the extraction yield by phenol- sulfuric acid method. Vc standard for comparison, study on antioxidants ability of polysaccharide in Trapa manshurica Flerow by reducing experiment, DPPH radical scavenging experiments, ABTS radical scavenging experiments.2. Study on extraction and antioxidants ability of flavonoids in Trapa manshurica Flerow.Single factor experiments were conducted to investigate the effects of operating parameters on the extraction by inner ebullition method. The following operating parameters were selected: desorption time, desorption concentration, desorption amount, extraction time, extraction temperature and extraction amount. Vc standard for comparison, study on antioxidants ability of Trapa manshurica Flerow extracts in Trapa manshurica Flerow by methods of potassium ferricyanide, DPPH and ABTS. And explored the synergistic antioxidant ability of Trapa manshurica Flerow extracts with Vc mixture.3. Preparation and antioxidants ability of gallic acid compoundsGallic acid-cisplatin complex was prepared by gallic acid and cisplatin. Gallic acid for comparison, study on antioxidants ability of gallic acid-cisplatin complex by reducing experiment, DPPH radical scavenging experiments, ABTS radical scavenging experiments. Result:1.Study on extraction and antioxidants ability of polysaccharide in Trapa manshurica Flerow.The optimized technology conditions was material liquid ratio 1:20, extraction temperature 80 ℃, extraction time 2 h. The extraction ratio of polysaccharide in Trapa manshurica Flerow was 30.81 %. The half clearance concentration on DPPH free radical was 1.37 mg/m L, the half clearance concentration on ABTS free radical was 1.84 mg/m L. The reducibility of polysaccharide in Trapa manshurica Flerow was weaker than Vc, and the ability of remove DPPH free radical and ABTS free radical were weaker than Vc.2.Study on extraction and antioxidants ability of flavonoids in Trapa manshurica Flerow.The optimized technology conditions was desorption time 20 min, desorption concentration 80 %,desorption amount 7 m L, extraction time 9 min,extraction temperature 90 ℃,extraction amount 50 m L. Under this optimized technology conditions, the content of total flavonoids in Trapa manshurica Flerow was 725.40 mg/100 g. The half clearance concentration of Vc and Trapa manshurica Flerow extracts on DPPH free radical were 56.50 μg/m L,64.18 μg/m L, after Vc combine with Trapa manshurica Flerow extracts IC50=54.73 μg/m L. The half clearance concentration of Vc and Trapa manshurica Flerow extracts on ABTS free radical were 69.61 μg/m L, 71.36 μg/m L, after Vc combine with Trapa manshurica Flerow extracts IC50=59.34 μg/m L. The reducibility from big to small was Vc and Trapa manshurica Flerow extracts compounds > Vc standard > Trapa manshurica Flerow extracts.3.Preparation and antioxidants ability of gallic acid compoundsGallic acid-cisplatin complex was prepared. The half clearance concentration of gallic acid and Gallic acid-cisplatin complex on DPPH free radical were 16.04 μg/m L, 62.02 μg/m L, the half clearance concentration on ABTS free radical were 32.58 μg/m L, 93.80 μg/m L, The reducibility of gallic acid was stronger than Gallic acidcisplatin complex. Conclusion:The optimized technology conditions of flavonoids and polysaccharides in Trapa manshurica Flerow were determined. The results show that,the antioxidants ability of polysaccharides was weaker than Vc, and the antioxidants ability of Trapa manshurica Flerow extracts was slightly lower than Vc. The antioxidants ability was enhanced obviously after Vc combine with Trapa manshurica Flerow extracts. The antioxidants ability of Gallic acid-cisplatin complex was obvious and it was provided a new ideas to reduce side effects of cisplatin. |
