Metabonomic Study On The Toxicity And Compatibility Of Vitamin C Yinqiao Tablets

Vitamin C Yinqiao Tablets is a kind of Compound preparation of Chinese and Western Medicine. Its toxicity and compatibility are studied in this subject in order to better explain the safety of vitamin C Yinqiao Tablets.First, the main toxic substances in vitamin C Yinqiao Tablets were conjectured through the literature screening. Chlorphenamine maleate and acetaminophen was identified as the main toxic substances in vitamin C Yinqiao Tablets.Analysis of rat urine by NMR, testing of some biochemical indicator of the rat and etc can be used to study effect on metabolism of male Wistar rats which caused by acetaminophen, chlorpheniramine maleate and a combination of both. The result suggest that the high dose group of the combination of acetaminophen and chlorpheniramine maleate(15.23 times as normal dose of medication of Vitamin C Yinqiao Tablet) show a strong side effects on body weight of rats, biochemical indicator, hepatic tissue sections and metabolite change. It was verified that combined use of acetaminophen and chlorpheniramine maleate is the main reason of vitamin C Yinqiao Tablets for the toxicity.According to the production process, the experiment is divided into the following groups:vitamin C, volatile oil(the extract of Forsythia suspensa,Lonicera japonica and Nepeta cataria L.), filtrate 1(the common extract was produced from Lophatherum gracile, semen sojae praeparatum, reed rhizome, Platycodon grandiflorus,Glycyrrhiza uralensis Fisch and the drug residues of volatile oil), filtrate 2(the extract of Fructus arctii), peppermint oil, Vitamin C Yinqiao Tablet(made in the laboratory), purchased Vitamin C Yinqiao Tablet. Each group(dose groups were 15.23 times the dose of Yinqiao Tablets normal medication) combined with high doses of the combination of acetaminophen and chlorphenamine maleate, respectively. Then metabolomics experiments was carried out. Through the test of body weight, biochemical indexes and metabolites in rats, it was found that all the groups had some effects on the toxicity of the combination of acetaminophen and chlorpheniramine maleate. Detoxify of vitamin C was not obvious.All data of the filtrate 1 group comes closer to the control group, so the detoxify of filtrate 1 is the best.The toxicity of the purchased Vitamin C Yinqiao Tablet is less than the combination of acetaminophen and chlorpheniraminemaleate. It suggested that the extract of light bamboo, fermented soybean, reed rhizome, radix platycodonis, licorice,forsythia thunbvahl, honeysuckle and Schizonepeta can reduce toxicity.Vitamin C Yinqiao Tablet is a classical compound preparation of Chinese and Western Medicine.Acetaminophen(antipyretic), chlorpheniramine maleate(anti allergic) is the commonly used anti cold Western medicine. The traditional Chinese herb medicine in Vitamin C Yinqiao Tablet has clearing away heat and toxic material, dispelling wind and heat, opening the inhibited lung-energy and help produce saliva and slake thirst base on the theory of traditional Chinese medical science. So the combination of both can increase the therapeutic effect and reduce toxicity of western medicine. The dose of all the experimental groups were 15.23 times as much as vitamin C Yinqiao Tablets normal dose. Under this dose, the data of filtrate 1 group comes close to the blank group.Results confirmed that clinical dose of vitamin C Yinqiao tablet was safe.
