Objective:To research the recombinant immunomodulatory protein of Ganoderma Lucidumof on aging mice induced by D-galactose anti-aging and whitening effect.Methods:(1) Effect of recombinant immunomodulatory protein of Ganoderm Lucid um on Anti-aging.(1) Experiments on animals:In addition to the normal control group, the rest of the four groups are induced aging model mice by D-galactose. The mice were randomly divided into 5 groups: normal control group, model control group,r LZ-8-low, medium and high dose group(from low to high doses respectively 660,990 and 1320 ug/kg). After the anesthesia obtain specimens for serological and histomorphology detection.(2) Experiments on cells : Cultivating human skin fibroblasts, divided the cells into four groups, collected cell culture supernatant at 24 h,48h, 72 h to detect collagen, hyaluronic acid and hydroxyproline content.(2) Effect of recombinant immunomodulatory protein of Ganoderm Luci dum on Whitening researchCultivting melanoma cells and Vaccinated in 6 holes, absorbed nutrient solution after 24 h and Joined the serum-free culture medium. Adding the r LZ-8 and made the final concentration of 8,16,24,32,48 ug/ml, and respectively within 24 h, 36 h, 48 h and 60 h after collecting cells and culture supernatant, detection of LDH activity. At the same time using thiazole blue colorimetry(determined by MTT method)determination of melanoma cell proliferation inhibition rate.Results:(1) When giving the drug, the mice began to lively, Quick action and the appearance improved in a certain degree.(2) Compared with the control group, the model group rat’s skin water were reduced in some degree after giving drug, but the results have no difference(P>0.05).(3) The results of hematoxylin-eosin staining showed: the mice of model group epidermis skin obviously thinning, dermis Collagen fiber began to more thinner, Skin adnexal presents degeneration and atrophy. At the same time, Dosage groups of mice showed better experimental results.(4) The results of Immunohistochemical staining showed: the content of model group I, III collagen and vimentin were all reduced. After giving drug, the content of these indicators are has a tendency of increase.(5) The results of serological and skin tissue homogenate showed: compared with the control group, SOD,CAT,GSH-Px,HYP and MT in the model group were significantly increased(P<0.05), However, the content of MDA Showed the opposite trend. After giving the drug, the activity of antioxidant enzymes have obviously improved, lipid peroxide content had significantly reduced.(6) The result of cell experiment show: compared with the control group, Each group of human skin fibroblasts I, III collagen, hyaluronic acid(HA) and hydroxyproline(HYP) levels were raised(P<0.05).(7) Compared with the control group, the proliferation rate of melanoma cell in the drug group were all slowly, at the same time, the activity of LDH and TYR were reduced too.Conclusion:(1) Recombinant immunoregulatory protein of ganoderma lucidum immuno modulatory protein can improve the appearance characteristics of aging mice, r aise the skin moisture content. At the same time, the content of model group I,III collagen and vimentin were all reduced. After giving drug, the content of these indicators are has a tendency of increase.The animal and cell experimentresults can fully proved that the effect of Ganoderma lucidum protein on antiaging.(2) Recombinant immunoregulatory protein of ganoderma lucidum immuno modulatory protein can improve the activity of SOD,CAT,GSH-Px,HYP and MT,reduce the lipid peroxide of MDA.It can improve the antioxidant ability of m ice.(3) In addition to this conclusion, Recombinant immunoregulatory protein of ganoderma lucidum immunomodulatory can also inhibit melanoma cell prolif eration, improve the activity of TYR, and confirmed the effect of whitening sk in.
