Objective: To investigate the relationship between endothelial nitric oxide synthase gene T-786 C,G-894T(eNOS T-786 C, eNOS G-894T)polymorphism and atrial fibrillation(AF) in Uyghur patients in Xinjiang.Methods: 100 cases of Uyghur patients with non valvular atrial fibrillation(NVAF) were selected as AF case group, and 100 cases of health patients as the control group. DNA samples were extracted from the peripheral blood. Polymerase chain reaction(PCR) method and DNA sequencing were used to detect the polymorphism. The mutation site was localized and identified. Results:The diameter of left atrial, total cholesterol, and low-density lipoprotein(LDL-C) in AF group was significantly higher than that in control group(P< 0.001). eNOS T-786 C,eNOS G-894 T polymorphism genotypes in AF group and control group sample were in the balance of Hardy-Weinberg. For eNOS gene T786 C, TT, TC CC genotype frequency in atrial fibrillation group were 0.905(181/200)、0.085(17/200) and 0.010(2/200). In the control group they were 0.870(174/200)、0.110(22/200) and 0.020(4/200), respectively.Genotype distribution of the two groups have no statistical significance(P>0.05). There was no significant difference between the C allele frequency(0.053) and the control group(0.075)(P>0.05). For eNOS gene G-894 T, GG, GT TT genotype frequency in atrial fibrillation group were 0.735(147/200), 0.225(45/200) and 0.040(8/200). In the control group they were 0.910(182/200), 0.080(16/200) and 0.010(2/200), respectively.Genotype distribution of the two groups have statistical significance(P<0.001). There was significant difference between the T allele frequency(0.152) and the control group(0.050)(P<0.001). In AF group, The distribution of eNOS G894 T genotype showedsignificant difference between Han and Uygur patients with atrial fibrillation(P<0.001).In AF group Uygur patients with atrial fibrillation, the frequency of T allele(0.240) and Han patients(0.065) showed statistical difference(P<0.001). Logistic regression analysis suggested that eNOS G894 T the G/T mutation, left atrial enlargement, total cholesterol abnormalities were the risk factors for non-valvular atrial fibrillation.Conclusions: The incidence of atrial fibrillation in Xinjiang Uygur had no significant correlation with eNOS T-786 C polymorphism. eNOS G-894 T polymorphism had correlation with the incidence of atrial fibrillation. In atrial fibrillation group, the eNOS G-894 T polymorphism showed significant difference(P<0.001) between the Han and Uighur.Genetic polymorphism may increase genetic susceptibility of non-valvular atrial fibrillation in Xinjiang Uighur.
