ObjectiveExplore the frequency of CDO-1 gene promoter mehtylation in serum and tumor tissue of patients with human brain glioma and to explore the effect of 5- Aza- 2 ’-deoxycytidine in U251.Methodsserum and tumor tissue of 50 patients suffered glioma were selected as experimental group. Necrotic brain tissue and serum of 20 patients with traumatic brain injury and serum of 10 healthy people were selected as control group. By using nest PCR and bisulifte PCR, we investigated methylation status of CDO-1 gene promoter in serum and tumor or necrotic brain tissue of both groups and their association with patient clinical history. We detected the methylation status of COD-1 gene promoter in U251 cultured in vitro with demethylating agent 5- Aza- 2 ’- deoxycytidine.ResultsWe found the frequency of CDO-1 gene promoter mehtylation among 44.0% of total tumor samples(22/50),with 64.0%(16/25) among female patients while 24.0%(6/25)among male patients, which was statistically significant, while similar results found in serum, 52.0%(13/25) in female to 20%(5/25) in male and 36.0%(18/50) totally.However, methylation was not found in the control group. The present study have failed to found correlation between the frequency of CDO-1 gene promoter mehtylation and clinical data except gender gap. The methylation status of COD-1gene promoter has decreased in U251 cultured in vitro with demethylating agent 5-Aza- 2 ’- deoxycytidine.ConclusionWe found the frequency of CDO-1 gene promoter mehtylation associated with human brain glioma while the different frequency among female and male patients was statistically significant. Blood samples have a potential as diagnosis of glioma. The methylation status of COD-1 gene promoter has decreased in U251 cultured in vitro with demethylating agent 5- Aza- 2 ’- deoxycytidine.
