ObjectiveTo explore the methylation status of p16 gene promoter region in human brain gliomas, to explore the effect of 5- Aza- 2 ’- deoxycytidine in U251.MethodsBy using Methylation-Specific PCR, bisulfite sequencing PCR, we detected the methylation status of p16 gene in 48 human glioma tumor and blood samples, By using 5- Aza- 2 ’- deoxycytidine,we detected the methylation status of p16 gene and biological character in U251.ResultsWe found the rate of methylation of p16 gene in blood is 37.5%( 18/48) and tumor tissue is 43.8%(21/48). The difference of p16 gene methylation rate and patients, Clinical data was not statistically significant. After 5-AZA-2-deoxycytidine treatment,we found that the methylation status of p16 gene changed in U251, we also found the changes of biological character in U251.ConclusionOur study Show that the p16 gene methylation Associated with glioma, blood samples have a potential as diagnosis of glioma.After 5-AZA-2-deoxycytidine treatment in U251,the methylation status of p16 gene disappeared and biological character changed. The drug, s anti-tumor activity may be applicable for clinical treatment.
