Wolfberry generally is a kind of Solanaceae genus wolfberry plants. Ningxia goji (Lycium barbarumL. Var. Barbarum.) has long been considered as a medicine and Edible tonic.,which was reported playing an important role in anti-oxidation, anti-aging, enhance immunity, scavenging free radicals. With further research, more and more varieties of wolfberry attentions. Hei goji (Lycium ruthenicum.murr.),known as "soft gold", "anti-aging king ", has been more concerned about the larger controversy. Its antioxidant activity has attracted widespread attention.Huang goji (Lycium barbarumL. Var. Auranticarpum K. F. Ching var. Nov.), Lycium barbarum L. Var. Barbarum.variants, was identified as the Solanaceae genus wolfberry. Huang goji is cultivated as an ornamental plant. Current research on different kinds of Chinese wolfberry fruit polyphenol lack of systematic, which restricts the development of Chinese wolfberry polyphenol components. This study is divided into two partsThe first part, the comparison of different wolfberry polyphenol and antioxidant activity.According to the literature, Chinese wolfberry was reported a good antioxidant activity. The total polyphenol, total flaconoid and total anthocyanin was measured in three kinds of wolfberry by enzyme labelling method. And reducing power and β-carotene oxidation inhibition of total polyphenol extracts three kinds was determined.28 kinds of polyphenol was identified in Lycium barbarumL. Var. Barbarum., which 15 kinds of polyphenol was found for the first time in the Ningxia goji.27 kinds of polyphenol was identified in Huang goji, wherein the compound is structurally similar to Ningxia goji, wherein the polyphenol components were first identified in Huang goji.26 kinds of polyphenol was identified in the Hei goji, which 22 species are found in the black for the first time in the Chinese wolfberry.Hei goji were better than the other two wolfberry in the total polyphenols, total flavonoids, total anthocyanins, total reducing power, and β-carotene oxidation inhibition rate.Hei goji can be developed as functional food and medicines because of the antioxidant activity.Second,Hei goji antioxidant activity evaluation and spermidine component characterizationChemical method to evaluating the antioxidant activity can not get the inaccurate results, because of the disability to simulate the body’s metabolic processes. In this study, Caco-2 cells as cell lines, in vitro oxidative damage by H2O2 simulated by measuring the intracellular ROS, IL-8, GSH content, to evaluate the antioxidant capacity of black wolfberry polyphenol extract. The results showed that Hei goji can effectively reduce the Caco-2 cells under oxidative stress in ROS, IL-8, GSH content in a dose-dependent manner, from the cellular level to prove that black wolfberry extracts (LRE) can significantly enhance the antioxidant capacity of Caco-2 cells. Hei goji contains a lot of polyphenols, and has strong antioxidant activity, it can be widely used in nutraceutical, pharmaceutical, cosmetics and a variety of natural products.To further confirm spermine compound discovered in black wolfberry, three kinds of phenolic amide was purified and identified by NMR and precise structure. Composition 1,2,3 were first discovered and identified from black wolfberry. Further determine the composition of a black Chinese wolfberry content and antioxidant activity. Studies have shown that (Dihydrocaffeoyl) caffeoyl spermidine has good antioxidant activity and other physiological activities. Further research is needed in the animals.
