Objective:To investigate the relationship between Uygur and Han plasma homocysteine (Hcy) and methylenetetrahydrofolate reductase (MTHFR) gene polymorphism and essential hypertension(EH) in Xinjiang. Methods:A case-control study was conducted by genotyping the representative variation in 598 hypertensive individuals including 286 Uyghur and 312 Han patients and 576 normotensive individual sincluding 270 Uyghur ethnic and 306 Han ethnic. The C677T genetic polymorphisms of the MTHFR enzyme were detected in all subjects by polymerase chain reaction-Restriction fragment length polymorphism (PCR-RFLP) analysis and the A1298C were detected by specific primer PCR and then measure its gene sequnce.Results:(1) Compared with the control, plasma Hcy levels were significantly higher in EH group in both Uyghur and Han ethnic(P<0.05). (2) The distribution of genotypes were in line with Hardy-Weinberg genetic equilibrium equation (P>0.05) in the Uygur and Han people. (3) There were significant differences in MTHFR C677T single nucleotide polymorphism (SNP) between EH patients and controls in both Uyghur and Han ethic (Uyghur:x2=11.592, P<0.05; Han:x2=10.471, P<0.05). However, the MTHFR A1298C has no statistical difference between EH patients and controls in both Uyghur and Han ethic (P>0.05). (4) Plasma levels of Hcy in 677 TT genotype were statistically higher than CC genotype (P<0.05). (5) After adjusting confounding factors such as gender, the logistic regression analysis indicated age (OR=1.018,95%CI=0.999-1.038 P<0.001),obesity (OR=2.504,95%CI=1.632-3.843, P<0.001), smoking (OR=4.574, 95%CI=2.483-8.424,P<0.001), drinking (OR=3.728,95%CI=1.944-7.150,P<0.001), hyperhomocysteine (OR=1.647,95%CI=1.068-2.539, P=0.024),677TT (OR=1.480, 95%CI=1.015-2.143, P=0.041) were independent risk factors for Uygur patients with EH, while age (OR=1.082,95%CI=1.034-1.132, P=0.001), obesity (OR=1.845, 95%CI=1.132-3.005, P=0.014), smoking (OR=3.023,95%CI=1.205-7.585, P=0.018), drinking (OR=5.459,95%CI=1.727-17.253, P=0.04), hyperhomocysteine (OR=2.725, 95%CI=1.737-4.276, P<0.001),677TT (OR=1.995,95%CI=1.287-3.093, P=0.02) were independent risk factors for EH patients in Han ethic. Conclusion:Age, obesity, smoking, drinking, hyperhomocysteine, MTHFR 677TT gene are independent risk factors for hypertention in both Uyghur and Han ethnic. No correlation was found existed between the gene polymorphism of MTHFRA1298C and EH patients of Uygur and Han nationality.
