Expression And Intratumoral Heterogeneity Of LN-5γ2 In Esophageal Squamous Cell Carcinoma

Objective:The present study aimed to evaluate clinical relevance and intratumoral heterogeneity of LN-5y2 expression in ESCC in order to explore its impact on the progression of ESCC and the potential for its application to the therapy of the disease.Methods:LN-5y2 protein was examined in 135 ESCC cases with four regions for each by immunohistochemistry andcorrelation with clinical pathological parameters was analyzed by statistics.Results:LN-5y2 protein moderate and high expression was detected in 40.0% (54/135) of tumor tissues but in none of morphologically normal operative margin tissues. Immunostaining was observed in 31.1% (23/74) of early stages (stages Ⅰ/Ⅱ) and 50.8% (31/61) of late stage (stage III) cases, with significant difference between these two groups (P= 0.023). There was no statistical association of LN-5y2 expression with age, sex of patients, PT sage, lymph node metastasis and differentiation degree of tumors. However, differential expression of LN-5y2 protein was found in different sampling sites of the same tumor, different carcinoma nests of the same sampling site, and different regions of the same carcinoma nest. The positive rate and location of LN-5y2 were different in esophageal squamous carcinoma. Thestrength and area of LN-5y2 expression were also different in Carcinomain situ,mild dysplasia,Moderate dysplasia and Serious dysplasia of esophageal squamous carcinoma.Conclusions:(1)The expression of LN-5y2 in esophageal squamous carcinoma exists heterogeneity. The heterogeneity expression exists between different regions in the same tumor tissue and different cases. The location of LN-5y2 in esophageal squamous carcinoma cases was different from tumor margin to nucleus and cytoplasm. The heterogeneity expression of LN-5y2 was diferent in different hyperplasia esophageal squamous carcinoma and the positive rate was diferent too.(2) The expression of LN-5y2 in esophageal squamous carcinoma was related to the clinical stages,however, has nothing to do with other pathological parameters. The experimentwas providing morphological evidence for targeted therapy and the study of esophageal squamous carcinoma.
