Objective To observe the effects of pigment epithelium-derived factor(PEDF) on the migration and invasion of human lung adenocarcinoma A549 cell, as well as the expression of metastatic-related proteins including fibronectin(FN) and matrix metalloproteinase 9(MMP-9).Methods A549 cells cultured without PEDF were defined as negative control gr oup, the cells in PEDF intervention group were added 720, 360 and 180 nmol/L PEDF, respectively. The effects of PEDF on the migration and invasion of A549 cell were detected by Transwell assay. The effects of PEDF on the expression of FN and MMP-9 m RNA were detected by semi-quantitative RT-PCR assay, m eanwhile the protein of them in supernatant of cell culture and within cells were detected by ELISA.Results The numbers of migrating cells in 720, 360, 180 nmol/L PEDF intervention groups and negative control group were(16.44±3.47),(30.78±2.17),(42.11±2.36),(63.67±1.45) and the numbers of invasive cells were(11.44±1.58),(21.67±1.20),(35.11±2.71),(44.89±1.07), respectively. Compared with the negative control group, the numbers of migrating cells and invasive cells in PEDF intervention groups were significantly lower(P<0.05). With the increase of PEDF concentration, the number of migrating and invasive cells decreased by degrees(P<0.05). The relative expression levels of FN m RNA of A549 cells in 720, 360, 180 nmol/L PEDF intervention groups and negative control group were(0.245±0.016),(0.378±0.021),(0.488±0.040) and (0.616±0.026), respectively. MMP-9 m RNA relative expression levels were(0.378±0.020),(0.521±0.047),(0.711±0.016) and(0.859±0.021).Compared with the negative control group, the expression levels of FN and MMP-9 m RNA of A549 cells in PEDF intervention groups were significantly lower(P<0.05) and the expression levels were remarkably decreased by degrees with the increase of PEDF intervention doses(P<0.05). The concentrations of FN protein in supernatant of cell culture in 720, 360, 180 nmol/L PEDF intervention groups and negative control group were(86.660±2.250),(104.789±1.314),(126.778±0.386),(146.944±3.396)ng/ml and within cells were( 108.700±3.349),( 124.389±2.760),( 133.822±4.047),(143.933±1.633)ng/ml, respectively. The concentrations of MMP-9 protein in supernatant of cell culture in each group were(316.311±1.831),(357.989±3.236),(373.300±2.250),(416.956±1.289)ng/ml and within cells were(311.567±4.206),(362.333±2.883),(391.278±1.680),(473.044±1.745)ng/ml. Compared with the negative control group, the concentrations of FN protein of A549 cells in PEDF intervention groups were significantly lower(P<0.05) and FN protein concentrations were remarkably decreased by degrees with the increase of PEDF intervention doses(P<0.05). Similarly, compared with the negative control group, the concentrations of MMP-9 protein of A549 cells in PEDF intervention groups were significantly lower(P<0.05) and MMP-9 protein concentrations were remarkably decreased by degrees with the increase of PEDF intervention doses(P<0.05).Conclusions PEDF can significantly inhibit the migration and invasion of A549 cells in vitro, furthermore, PEDF can significantly inhibit the expression of metastasis associated proteins(FN and MMP-9).
