Objective In recent years, the researches carried out in the domestic and foreign people indicated that HLA-A*3001, HLA-A*3101, HLA-B*1502, HLA-B*38, HL A-B*5801 alleles may be associated with lamotrigine-induced cutaneous adverse reactions,but there is no report of this kind of research in Xinjiang Uygur. So in this study, we aim to investigate the relationship between HLA-A*3001,HLAA*3101,HLA-B*1502,HLA-B*38,HLA-B*5801 alleles and lamotrigine induced cut aneous adverse reactions in Xin Jiang Uyghur epilepsy patients.Methods HLA-A*3001、HLA-A*3101 、HLA-B*1502、HLA-B*38、HLA-B*5801alleles genotyping was performed by a polymerase chain reaction-sequence speci fic primers(PCR-SSP) method in 28 subjects who had lamotrigine.(10 Uyghur s ubjects had lamotrigine induced MPE,18 lamotrigine tolerant controls),and 30 hea lthy individuals. Statistical analyses were performed using SPSS version 19.0(SP SS, Chicago, IL, USA), Corrected Chi-square test was used to explore the differe nces in frequency of the HLA allele among the groups,and independent t test w as used for continuous variables. The level of statistical significance was set at P< 0.05(two-sided). Odds ratios(ORs) with corresponding 95% confidence interv als(CIs) were calculated to identify the association between HLA alleles and LT G induced cutaneous adverse reactions.Results There seemed to be no significant differences in the positive frequency of subjects with the HLA-A*3001(P=0.936, OR=2, 95%CI 0.236-16.928), HLAA*3101(P=0.585, OR=4.25, 95%CI 0.334-54.066),HLA-B*1502(P=0.875, OR=0.818, 95%CI 0.066-10.117), HLA-B*38(P=0.875, OR=0.818, 95%CI 0.066-10.117),HLA-B*5801(P=0.585, OR=4.25, 95%CI 0.334-54.066) alleles between the LTG-MPE group and LTG-tolerant group, and there was also no significant difference s in the positive frequency of subjects with the HLA-A*3001(P=0.543, OR=3.5,95%CI 0.424-28.915), HLA-A*3101(P=0.783, OR=2.25, 95%CI 0.318-15.901),H LA-B*1502(P=0.811, OR=1.364, 95%CI 0.112-16.577), HLA-B*38(P=0.893, OR=0.491, 95%CI 0.051-4.698), HLA-B*5801(P=0.298, OR=7.25, 95%CI 0.581-90.545) alleles between the LTG-MPE group and healthy volunteers..Conclusion HLA-A*3001 and other HLA alleles are not directly associated with LTG-MPE in Xin Jiang Uyghur epilepsy patients,but its small sample size limit s the power of text, so more researches with bigger sample size are needed for further prove.
