Scutellarin, is the main effective components of Erigeron breviscapus, and its quality control index component. According to reports, scutellarin can penetrate the blood-brain barrier, can significantly improve the cerebral blood circulation, for the prevention and treatment of cardiovascular and cerebrovascular diseases, good curative effect, is a very high medicinal value of plant raw materials.Scutellarin oral preparation on the market a lot, but there are still poor purity, oral absorption effect is not good problem. The aim of this paper is to improve the purity and the effect of oral absorption, and to study the preparation technology of high purity and multi crystal, including the following 3 aspects:(1)Purification of research:In the preparation process of previous reports on the basis of process optimization, the introduction of washing water, acetone solution ratio and other parameters on the purity and yield of. Ultimately determine scutellarin purification process:with distilled water into 7 times the volume of the solution(M/V), water bath heating, adjust the solution to p H 7.0, crystallization temperature of 20 DEG C, stirring speed 30 rpm, stirring time 16 h, filtration, and the cake with 12 times the amount of distilled water flushing and vacuum drying, solid powder. Take a certain amount of powder, in 80% acetone solution to adjust p H to acid, stirring, static, vacuum drying, get the content of 98.7% of the B, yield of66.3%.(2)Polymorphs of preparation: Take self-made purity >98% scutellarin amount,dissolved in different solvents, crystallization, according to the characteristics of the microscope, IR, DSC, TGA, and finally get the four different crystal type. Among them, 1kind is mixed crystal type, two is that have not been reported.(3)Polymorphs oral absorption effect evaluation: By measuring the crystal type in p H2.0-7.8 table view of oil-water partition coefficient of different simulation of breviscapine B prime solubility in the organisms of the gastrointestinal tract that preliminary evaluation: in methanol scutellarin new crystalline polymorph, has good oral absorption.
